We present a genome assembly from an individual female (the Plain Longtail butterfly; Arthropoda; Insecta; Lepidoptera; Hesperiidae). The genome sequence is 610.1 megabases in span. Most of the assembly is scaffolded into 32 chromosomal pseudomolecules, including the Z and W sex chromosomes. The mitochondrial genome has also been assembled and is 15.54 kilobases in length. Gene annotation of this assembly on Ensembl identified 18,506 protein coding genes.

We lack tools to edit DNA sequences at scales necessary to study 99% of the human genome that is noncoding. To address this gap, we applied CRISPR prime editing to insert recombination handles into repetitive sequences, up to 1697 per cell line, which enables generating large-scale deletions, inversions, translocations, and circular DNA. Recombinase induction produced more than 100 stochastic megabase-sized rearrangements in each cell.
