Cordycepin, a nucleoside analog, is widely used in medicine and health products. However, the production of cordycepin from Cordyceps militaris faces the challenges of low productivity and high rate of greenhouse gas emissions. In this study, by optimizing the cordycepin biosynthesis pathway through promoter combination, Kozak sequence, and enzyme fusion, enhancing the methanol assimilation capacity in peroxisomes, adjusting the synthesis of NADPH and ATP, and combining the enhanced supply of adenosine and 3'-AMP, the cordycepin high-yield strain Pp29 was constructed, which produced 1551.44 mg/L cordycepin by shake-flask fermentation. In fed-batch fermentation, Pp29 achieved the highest yield (8.11 g/L, 67.64 mg/g DCW, and 1.35 g/L/d) to date in 10 L fermenter, and the CO emissions were 1.9-17.3 times lower than C. militaris and other yeast systems. This study provide basis for Pichia pastoris to be used as chassis cell for synthesizing cordycepin and other nucleoside analogs by methanol as carbon source.

The methylotrophic yeast Pichia pastoris (also known as Komagataella pastoris) is an ideal host for producing proteins and natural products. Enhancing homologous recombination (HR) is helpful for improving the precision of genome editing, but results in stress to cellular fitness and is harmful for industrial applications. To overcome these challenges, we developed a tetracycline repressor protein (TetR)/tetO2 inducible system to dynamically regulate the HR-related gene RAD52 in P.

Apostichopus japonicus is a crucial aquaculture species in Asia, and seedling production is fundamental to its cultivation. Nevertheless, the reproductive behavior of this species remains difficult to regulate artificially. Although the spawning mode of sea cucumbers is well documented, the endocrine mechanisms that regulate the synchronized processes of oocyte maturation and spawning behavior remain poorly understood.

This study reports methods for reductive microbial deuteration with a high degree of isotopic labelling, focusing on economical approaches using various yeast strains and inexpensive carbon sources. A strategy of α-hydrogen exchange followed by deuterative microbial reduction gave >95% backbone perdeuteration for crucial chiral building blocks for medicinal and analytical applications, without loss of the enantioselectivity demonstrated in the corresponding protiative processes. Under an air atmosphere, strain MBG5177 outperformed baker's yeast in the synthesis of (2-H)solketal, while strain X-33 provided >95% deuteration in the synthesis of the two studied building blocks using methanol- as an affordable carbon source.

Xylitol, known for its health benefits, is a valuable compound in the food and pharmaceutical industries. However, conventional chemical production methods are often unsustainable for large-scale applications, prompting the need for alternative approaches. This study demonstrates a significant enhancement in xylitol production using microbial cell factories, optimized through metabolic engineering.

As a promising osmolyte, ectoine has been widely applied in cosmetics, food, and pharmaceutical industries in recent years, therefore its biomanufacturer has attracted increasing interest. Ectoine-producing isolates were previously screened from halophilic microorganisms. After ectoine synthetase was identified, genetic engineering of Escherichia coli, Corynebacterium glutamicum, and Hansenula polymorpha were employed to produce ectoine.
