Quantum chemical investigation of electronic transitions of mitorubrin azaphilones.

Fungal azaphilones are a broad class of naturally-occurring pigments with diverse applications. Among the azaphilone pigments, mitorubrins are well recognized for their antiviral, antibacterial, antifungal, antiprotozoal, antidiabetic, and antiaging activities in addition to their well-known yellow-orange color. This makes these pigments interesting candidates for use in foods, as cosmetics, and as medicines. In particular, if it is desired to modify the properties of mitorubrin-based pigments, for example by derivatization, it is essential to have an understanding of the electronic spectra of the parent molecules. We have therefore undertaken a computational study of a series of mitorubrins, comparing our computed results with experimental UV/visible spectra. Both density-functional theory (DFT) and coupled-cluster (CC2) methods have been used, and in general, the results are in very good agreement with observation. In order to provide a simple and useful picture of the spectra we analyze the stronger transitions in terms of natural transition orbitals (NTOs).

One new Sesquiterpene wikstromicrol (1), one new lignan (7R, 8S, 8'S)-9-acetoxyl (-)-isolariciresinol (8), and twelve known compounds were isolated from the branches and leaves of Wikstroemia micrantha. The structures of new compounds were elucidated by extensive interpretation of spectroscopic data and quantum chemical calculations of ECD spectra. The bioactivity assay showed that compounds 6 and 7 had weak cytotoxicity against MCF-7, HCT-116, and HL-60 cell lines with IC50 values ranging from 33.

The History of Studies on Oxetane Ring Formation in Paclitaxel Biosynthesis.

There is no doubt that breakthroughs in the enzyme-mediated formation of the oxetane ring in paclitaxel biosynthesis constitute significant milestones in the biosynthesis of complex natural products. In this review, we summarize the understanding of the biosynthesis of the oxetane ring of paclitaxel from different viewpoints. Generally, it covers five aspects, (1) a different understanding of the mechanistic formation of the oxetane ring on the basis of sound chemical reasoning, (2) a reasonable speculation of the biosynthetic pathways and suitable surrogate substrates for oxetane ring formation based on the natural and chemical logical analysis, (3) Taxus genome-enabled enzymes identification, (4) the discovery of different enzymes that mediate oxetane ring formation, and (5) a mechanistic investigation involving the use of isotopic labelling experiments and quantum chemical calculations.

Ultrasensitive point-of-care multiplex diagnosis for influenza virus based robust quantum dot microsphere-lateral flow immunoassay.

Influenza A virus (IAV) and influenza B virus (IBV) with similar symptoms of infection caused a serious disease burden and economic losses in annual epidemic season, so it is important to quickly and accurately detect and distinguish between IAV and IBV during influenza season. Herein, the quantum dot microspheres (QDMS) were synthesized and applied to lateral flow immunoassays (LFIA), and a point-of-care (POC) biosensor that can discriminately and simultaneously diagnose IAV and IBV within 10 min was established. A double-sandwich QDMS nanotags was synthesized by immobilizing hydrophobic quantum dots (QDs) with chemical bonding method on a silica sphere template with an outer silica shell protection showed excellent stability and high fluorescence.

Efflux pumps that transport antibacterial drugs out of bacterial cells have broad specificity, commonly leading to broad spectrum resistance and limiting treatment strategies for infections. It remains unclear how efflux pumps can maintain this broad spectrum specificity to diverse drug molecules while limiting the efflux of other cytoplasmic content. We have investigated the origins of this broad specificity using theoretical models informed by the experimentally determined structural and kinetic properties of efflux pumps.
