Pesticide use increases annually, and Brazil is the world's largest consumer. However, unlike the European Union (EU), there is no established limit value for pesticide mixtures in drinking water, and therefore the concentration of pesticides can reach 3354 times the EU limit. Thus, determining the risk of exposure to pesticide mixtures and their main metabolites is challenging and requires the use of alternative methods. In the present study, the Common Carp Brain (CCB) cell line was used to evaluate the in vitro toxicity of relevant pesticide mixtures (glyphosate, 2,4-D, atrazine, and mancozeb) and their main metabolites after 72 h of exposure. The tested concentrations were based on the Acceptable Daily Intake (ADI) defined by Brazilian legislation. The results showed that cells exposed to lower concentrations of the pesticide mixtures and the pesticide + metabolite mixtures were affected by a decrease in cell confluence, resazurin metabolism, and wound healing capacity. The IBR index showed that lower concentrations had more severe effects, suggesting the absence of safe concentrations of these pesticide and metabolite mixtures for the CCB cell line within the tested concentration range. These findings raise concerns about the effects of exposure to these substances on animal and human health.

The co-occurrence of pesticides in aquatic ecosystems highlights the need for studies investigating their potential toxicity as mixtures to the aquatic biota. Well-designed studies are essential to assess the presence and toxicity of relevant pesticide mixtures, particularly those such as the chloroacetamide herbicide metolachlor (MET), the triazole fungicide epoxiconazole (EP) and the diamide anthranilic insecticide chlorantraniliprole (CAP), which have not been previously tested, and whose co-occurrence is possible in waters close to cultivated areas. A solid phase extraction ultra-performance liquid chromatography-tandem quadrupole mass spectrometry method was developed to quantify equivalent toxicity concentrations for CAP, EP, and MET in artificial freshwater during acute toxicity tests.

Previous epidemiologic studies examining thyroid function and chemical exposures have typically focused on a single or a limited number of chemical classes, often neglecting the effects of chemical mixtures. This study addressed this gap by exploring the associations between exposure to hundreds of chemicals and thyroid function using an exposome-wide association study (ExWAS) approach and National Health and Nutrition Examination Survey (NHANES) data. We analyzed data from three NHANES cycles (2007-2008, 2009-2010, 2011-2012), which include measures of thyroid function (free and total triiodothyronine [T3], free and total thyroxine [T4], thyroid-stimulating hormone [TSH]) and chemical biomarker concentrations from 9,082 participants.
