25-Hydroxycholesterol modulates synaptic vesicle endocytosis at the mouse neuromuscular junction.

Laboratory of Biophysics of Synaptic Processes, Kazan Institute of Biochemistry and Biophysics, FRC Kazan Scientific Center of RAS, 2/31 Lobachevsky St, Kazan, 420111, RT, Russia.

Many synaptic vesicles undergo exocytosis in motor nerve terminals during neuromuscular communication. Endocytosis then recovers the synaptic vesicle pool and presynaptic membrane area. The kinetics of endocytosis may shape neuromuscular transmission, determining its long-term reliability.

Background: Alzheimer disease (AD) plasma biomarkers change in the preclinical stage of AD. However, the robustness of the discrimination performance of these biomarkers, as well as their association with longitudinal primary pathology (amyloid and tau) changes, is less understood. We aimed to determine the ability of baseline and longitudinal plasma amyloid-β (Aβ)42/40, p-tau181, GFAP and NfL to detect primary pathology in CU individuals at risk of AD.

Alzheimers Dement

December 2024

Department of Psychiatry and Neurochemistry, Institute of Neuroscience and Physiology, The Sahlgrenska Academy, University of Gothenburg, Mölndal, Gothenburg, Sweden.

Background: Emerging evidence underscores the importance of neuroinflammation in the progression of Alzheimer's disease (AD) pathophysiology. Recent studies indicate the involvement of the inflammatory mechanisms both in amyloid- β (Aβ) and tau deposition in the brain. Nevertheless, due to the complexity of the immune responses and the intricate interplay between the peripheral and the central nervous systems, identifying biomarkers that reflect the brain´s inflammatory state in AD has been a challenge.

Alzheimers Dement

December 2024

Clinical Memory Research Unit, Department of Clinical Sciences Malmö, Faculty of Medicine, Lund University, Lund, Sweden.

Background: Blood-based biomarkers have demonstrated great promise for identifying biomarker-confirmed Alzheimer's disease. We aimed to evaluate whether blood-based biomarkers could optimize the referral of memory clinic patients to a tau-PET exam, which is crucial for prognostic evaluation.

Method: The study measured various plasma biomarkers (Aβ42/Aβ40, pTau181, pTau217, pTau231, NfL, and GFAP) and compared them with tau-PET scan results in patients with subjective cognitive decline, mild cognitive impairment, or dementia.

Alzheimers Dement

December 2024

Zhongnan Hospital, Wuhan University, Wuhan, China.

Background: Vascular cognitive impairment (VCI), including vascular cognitive impairment-no dementia (VCIND) and vascular dementia, is a cognitive impairment syndrome caused by cerebrovascular disease and its risk factors. Among people over 60 years old, the prevalence of VCIND is about 15-20%. VCIND, as the early stage of VCI, has become a focus of current research due to the fact that its patients are at greater risk of developing dementia and can benefit greatly from early intervention.
