The ability to suppress inappropriate actions and respond rapidly to appropriate ones matures late in life, after puberty. We investigated the development of this capability in monkeys trained to look away from a lone, bright stimulus (antisaccade task). We evaluated behavioral performance and recorded neural activity in the prefrontal cortex both before and after the transition from puberty to adulthood. Compared to when young, adult monkeys processed the stimulus more rapidly, resisted more effectively the involuntary urge to look at it, and adhered to the task rule more consistently. The spatially selective visuomotor neurons in the prefrontal cortex provided neural correlates of these behavioral changes indicative of a faster transition from stimulus-driven (exogenous) to goal-driven (endogenous) control within the time course of each trial. The results reveal parallel signatures of cognitive maturation in behavior and prefrontal activity that are consistent with improvements in attentional allocation after adolescence.

In task-shared, mental health, and psychosocial support interventions, monitoring the quality of delivery (fidelity and competence) of nonspecialist providers is critical. Quality of delivery is frequently reported in brief, summary statistics, and while both fidelity and competence scores tend to be high, rarely have factors associated with quality of delivery in low-resource, mental health, and psychosocial support interventions been examined using inferential statistics. Understanding both modifiable and non-modifiable predictors of quality of delivery is important for adapting training and supervision approaches throughout intervention delivery.

The ForenSeq™ DNA Signature Prep kit has not been thoroughly tested with crude buccal swab lysates in large-scale population studies using massively parallel sequencing (MPS). Commonly used lysis buffers for swabs intending to undergo direct polymerase chain reaction (PCR) are SwabSolution™ and STR GO! Lysis Buffers, and these have been successfully used to generate population data using capillary electrophoresis (CE) systems. In this study, we investigated the performance and optimisation of SwabSolution™ and STR GO! lysates with the ForenSeq™ DNA Signature Prep workflow and addressed the challenge of failed MPS profiles in initial trials.

Reduced fidelity of these vacuolar organelles is increasingly being recognized as important in instigating cell-autonomous immune cell activation. We reasoned that exploring the role of BLOC1S1 in CD4 T cells, may further advance our understanding of regulatory events linked to mitochondrial and/or endolysosomal function in adaptive immunity.
