Mounting evidence supports the potential of dietary bioactives to reduce chronic disease risk. -trans-caffeoyltyramine (NCT) and -trans-feruloyltyramine (NFT) have been hypothesized to drive regulation of gut permeability, but these components have not yet been studied in the context of the human gut microbiome. This work examined whether purified NCT and NFT, or a hemp hull product containing NCT and NFT (Brightseed® Bio Gut Fiber™), can impact the gut microbiome using an fermentation assay. Representative human gut microbiomes were treated with Bio Gut Fiber™ or NCT and NFT and compared to starch and methylcellulose, as controls, . Stronger changes were exerted by Bio Gut Fiber™, NCT, and NFT. Communities treated with Bio Gut Fiber™ saw increased productivity and diversity. We found a dose-dependent effect of NCT and NFT on microbial communities. Here, we describe novel potential for hemp-derived bioactives to shape the gut microbiome.

Structure-based identification of HNF4α agonists: Rosmarinic acid as a promising candidate for NAFLD treatment.

Unlabelled: The prevention and treatment of metabolic disorders, such as non-alcoholic fatty liver disease (NAFLD), have emerged as critical global health challenges. Current lipid-lowering pharmacotherapies are associated with side effects, including hepatotoxicity, rhabdomyolysis, and decreased erythrocyte counts, underscoring the urgent need for safer therapeutic alternatives. Hepatocyte nuclear factor 4α (HNF4α) has been identified as a pivotal regulator of lipid metabolism, making it an attractive target for drug development.

Article Synopsis
  • Researchers discovered strong HNF4α agonists, NCT and NFT, that regulate fat storage in the liver by inducing lipophagy via dihydroceramides.
  • These agonists are structurally related to weak HNF4α activators and effectively cleared fat from cells in vitro.
  • In experiments with DIO mice, NCT improved liver function and reduced fat buildup, showing promise as a safe treatment for non-alcoholic fatty liver disease (NAFLD).

Background: The objective of this study was to make a quantitative comparison of flortaucipir PET retention with pathological tau and β-amyloid across a range of brain regions at autopsy.

Methods: Patients with dementia (two with clinical diagnosis of AD, one undetermined), nearing the end of life, underwent 20-min PET, beginning 80 min after an injection of ~370 mBq flortaucipir [F]. Neocortical, basal ganglia, and limbic tissue samples were obtained bilaterally from 19 regions at autopsy and subject-specific PET regions of interest corresponding to the 19 sampled target tissue regions in each hemisphere were hand drawn on the PET images.

A combination of autoradiography and horseradish peroxidase histochemistry was used to identify telencephalic structures linking the sensory and motor components of the trigeminal system in the pigeon. A direct telencephalic projection from the principal trigeminal sensory nucleus upon the nucleus basalis via the quintofrontal tract was confirmed. Nucleus basalis projects upon a belt of neurons within the overlying neostriatum.
