The effects of three glucocorticoids on random migration (RM) and oriented migration (OM) of dog blood leukocytes either from dogs treated in vivo (at therapeutic dosage regimen) or leukocytes treated in vitro (at pharmacological concentrations), were investigated using an agarose gel technique. After in vivo treatment, methylprednisolone sodium succinate (MPSS) produced a stimulation of both RM and OM in the three treated dogs. Dexamethasone produced a stimulation of both RM and OM in the three treated dogs. Dexamethasone produced a stimulation of these parameters in all but one of the three treated dogs, but the difference was not statistically significant. After in vitro treatment of leukocytes from eight dogs, MPSS significantly stimulated OM. Dexamethasone was without significant effect except at a higher than therapeutic concentration (0.5 micrograms/ml), for which RM was stimulated. Hydrocortisone sodium succinate was without statistically significant effect. Under no conditions, except after suprapharmacological concentrations, did glucocorticoids inhibit RM or OM.

The Rbfox proteins regulate alternative pre-mRNA splicing by binding to the RNA element GCAUG. In the nucleus, most of Rbfox is bound to the large assembly of splicing regulators (LASR), a complex of RNA-binding proteins that recognize additional RNA motifs. Here we established an Il17a-GFP:Ifng-YFP double-reporter mouse strain to analyze at unprecedented depth the transcriptomes of pure γδ cell versus γδ cell populations from peripheral lymph nodes. Within a very high fraction of differentially expressed genes, we identify a panel of 20 new signature genes in steady-state γδ cells versus γδ cells, which we further validate in models of experimental autoimmune encephalomyelitis and cerebral malaria, respectively.

Granulocyte colony stimulating factor (G-CSF) has been shown in mammalia to activate a series of signal transduction systems and exert various biological effects, such as controlling the differentiation, proliferation, and survival of granulocytes, promoting the movement of hematopoietic stem cells from the bone marrow to the bloodstream, and triggering the development of T cells, dendritic cells, and immune tolerance in transplants. In this study, the mRNA of flounder G-CSF (PoG-CSF) and its receptor (PoGCSFR) were detected and widely expressed in all examined tissues with the highest expression in peritoneal cells. G-CSF and GCSFR cells were observed to be abundantly distributed in the leukocytes from the peritoneal cavity, followed by head kidney.
