Lameness and restricted mobility are a significant concern in companion animals experiencing chronic pain, inflammation, or age-related pathologies. The growing awareness of health risks and side effects associated with the long-term use of prescription analgesics requires different management strategies to address these issues. In this study, we conducted a crossover evaluation of the effect of full spectrum hemp oil dosed orally at 2 mg/kg BID phytocannabinoids for 8 weeks in dogs ( = 37) living with pain in common household settings. Owner-reported canine pain, home activity, accelerometer-based activity, walkway-based gait, and tolerability were assessed at each phase of the study. Secondary endpoints included changes in blood biochemistry, liver enzymes, inflammatory biomarkers, and plasma metabolites. The intervention was positively associated with a decrease in pain scores (-46.2%, = 0.0016), increased ability to walk up and down the stairs (10.6-14.7%, < 0.05), and improved daily activity (25.9%, = 0.0038). Decreases in plasma levels of proinflammatory cytokines TNF-α, IL-6, and IL-8 were also observed. Taken together, these findings suggest that the benefits of nutritional supplementation with hemp oil could include control of pain, greater mobility, and an overall improvement in the animal wellbeing.

Background: The growing number of people living with Alzheimer's disease and related dementias (ADRD) has led to an increased interest in the experiences of informal caregivers. Effective instruments to measure both negative and positive aspects of caregiving and validated with diverse caregiver populations, are needed to inform the design and evaluation of targeted interventions. This study (a) reviews extant literature on instruments developed to measure the range of roles and experiences of unpaid caregivers of people living with ADRD, (b) describes characteristics of the populations used to validate these instruments, and (c) discusses the usefulness, applicability, and generalizability of current measures METHOD: A scoping review was performed following the methodological framework of Aromataris and Munn (2020).

Myelin abnormalities in white matter have been implicated in the pathophysiology of psychotic spectrum disorders (PSD), which are characterized by brain dysconnectivity as a core feature. Among evidence from in vivo MRI studies, diffusion imaging findings have largely supported disrupted white matter integrity in PSD; however, they are not specific to myelin changes. Using a multimodal imaging approach, the current study aimed to further delineate myelin and microstructural changes in the white matter of a young PSD cohort.

Breast cancer is a highly heterogeneous disease whose prognosis and treatment as defined by the expression of three receptors-oestrogen receptor (ER), progesterone receptor and human epidermal growth factor receptor 2 (HER2; encoded by ERBB2)-is insufficient to capture the full spectrum of clinical outcomes and therapeutic vulnerabilities. Previously, we demonstrated that transcriptional and genomic profiles define eleven integrative subtypes with distinct clinical outcomes, including four ER subtypes with increased risk of relapse decades after diagnosis. Here, to determine whether these subtypes reflect distinct evolutionary histories, interactions with the immune system and pathway dependencies, we established a meta-cohort of 1,828 breast tumours spanning pre-invasive, primary invasive and metastatic disease with whole-genome and transcriptome sequencing.

Non-close-packed crystalline arrays of colloidal particles in an elastic matrix exhibit mechanochromism. However, small interparticle distances often limit the range of reversible color shifts and reduce reflectivity during a blueshift. A straightforward, reproducible strategy using matrix swelling to increase interparticle distance and improve mechanochromic performance is presented.
