Inhalable dry powders of a monoclonal antibody against SARS-CoV-2 virus made by thin-film freeze-drying. | LitMetric

AI Article Synopsis

Article Abstract

Monoclonal antibodies (mAbs) represent a promising modality for the prevention and treatment of viral infections. For infections that initiate from the respiratory tract, direct administration of specific neutralizing mAbs into lungs has advantages over systemic injection of the same mAbs. Herein, using AUG-3387, a human-derived mAb with high affinity to the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) and its various variants, as a model mAb, we formulated the mAb into dry powders by thin-film freeze-drying, confirmed that the AUG-3387 mAb reconstituted from the dry powders retained their integrity, high affinity to the SARS-CoV-2 spike protein receptor binding domain (RBD), as well as ability to neutralize RBD-expressing pseudoviruses. Finally, we showed that one of the AUG-3387 mAb dry powders had desirable aerosol properties for pulmonary delivery into the lung. We concluded that thin-film freeze-drying represents a viable method to prepare inhalable powders of virus-neutralizing mAbs for pulmonary delivery into the lung.

This study introduces novel cospray-dried (Co-SD) formulations of simvastatin, a Nrf2 activator ROCK inhibitor, with l-carnitine as molecular mixtures in various molar ratios for targeted pulmonary inhalation aerosol delivery in pulmonary hypertension, optimized for excipient-free dry powder inhalers (DPIs). The two components were spray-dried at various molar ratios by using different starting feed solution concentrations and process parameters. In addition to comprehensive physicochemical characterization, in vitro aerosol dispersion performance as DPIs using two FDA-approved DPI devices with different shear stress properties, in vitro viability as a function of dose on 2D human pulmonary cellular monolayers and on 3D small airway epithelia human primary cultures at the air-liquid interface (ALI), and in vitro transepithelial electrical resistance (TEER) at the ALI were conducted.

A survey of physicians, biomedical researchers and college-educated adults in urban north India about inhaled therapies.

Tuberculosis (Edinb)

December 2024

CSIR-Central Drug Research Institute, Sector 10A, Janakipuram Extension, Lucknow, 226031, UP, India. Electronic address:

We surveyed 15 persons with a medical qualification, 133 graduate students doing biomedical research and 56 students or working people with a college education in any discipline. Questions were designed to gauge awareness about inhaled therapies for tuberculosis (TB), non-tubercular mycobacterial lung disease (NTM-LD) and idiopathic pulmonary fibrosis (IPF). Respondents from six cities in North India, aged between 21 and 57 years answered 20 questions.

The aim of this study was to analyse the effect of breathing parameters, age, gender and disease status on the lung doses of the two ICS+LABA fixed combination dry powder drugs. Breathing parameters of 113 COPD patients were measured while inhaling through emptied NEXThaler® and Ellipta® inhalers and the corresponding lung doses were calculated. Lung dose of Foster® NEXThaler® was superior to the lung dose of Relvar® Ellipta® in around 85% of the patients.

Medium-chain antimicrobial lipids are promising antiviral agents to inhibit membrane-enveloped viruses such as African swine fever virus (ASFV) and influenza A virus (IAV) in livestock applications. However, current uses are limited to feed pathogen mitigation due to low aqueous solubility and the development of water-dispersible lipid formulations is needed for broader application usage. In this study, we report a water-dispersible antimicrobial lipid mixture of monoglycerides and lactylates that can inhibit ASFV and IAV and exhibits antiviral properties in drinking water and feed matrices.

Free-standing bimetallic Co/Ni-MOF foams toward enhanced methane dry reforming under non-thermal plasma catalysis.

J Colloid Interface Sci

December 2024

Laboratory of Plasma and Energy Conversion, School of Physics and Optoelectronic Engineering, Ludong University, Yantai, China. Electronic address:

Understanding of the structure and interfacial merits that reactive metal-organic frameworks (MOFs) undergo is critical for constructing efficient catalysts for non-thermal plasma-assisted conversion of greenhouse gases. Herein, we proposed a free-standing bimetallic (Co/Ni) MOFs supported on bacterial cellulose (BC) foams (Co/Ni-MOF@BC) toward the coaxial dielectric barrier discharge (DBD) plasma-catalytic system, of which the Co/Ni ions coordination demonstrated an intriguing textual uplifting of the malleable BC nanofiber network with abundant pores up to micrometer-scale, which could impart a more intensive predominant filamentary microdischarge current to 180 mA with stronger plasma-catalytic interaction. Remarkably, compared to the monometallic MOF@BC foams, this bimetallic Co/Ni-MOF@BC also delivered a substantially improved alkaline absorption ability as further confirmed by the CO- temperature-programmed desorption (TPD) result.
