Thermo-responsive hydrogels can generate the actuation force through volumetric transitions in response to temperature changes. However, their weak mechanical properties and fragile actuation performance limit robust applications. Existing approaches to enhance these properties have typically depended on additional components, leading to an unavoidable interference to the actuation performance. In this work, robust thermo-responsive hydrogels are fabricated through solvent engineering. A particular solvent, N-methylformamide, interacts affinitively with the carbonyl group of N-isopropylacrylamide monomer, solubilizes the monomer with extremely high concentration, stabilizes chain propagation during polymerization, and greatly increases chain lengths and entanglements of the resulting polymer. The synthesized hydrogels are highly elastic, strong, and tough, displaying remarkable thermo-responsive contractile actuation. The simple synthetic process can broaden its applicability in designing robust functional hydrogel applications.

A clinical need exists for more effective intravitreal (IVT) drug delivery systems (DDS). This study tested the hypothesis that a novel biodegradable, injectable microsphere-hydrogel drug delivery system loaded with aflibercept (aflibercept-DDS) would exhibit long-term safety and biocompatibility in a non-human primate (NHP) model. This paper investigates the integration of thermo-responsive materials into bio-inspired structures, combining biomimicry and adaptive technologies in architecture. Adhesive epidermal hydrogel electrodes are essential for achieving robust signal transduction and cardiac arrhythmia diagnosis, but detachment of conventional adhesive dressings easily causes secondary damage to delicate wound tissues due to lack of programmable capability of changed adhesion. Herein, we developed hydrogel-based skin-interfacing electrodes capable of on-demand programmable adhesion and detachment to capture electrocardiogram signals for diagnosing cardiac arrhythmia. This was achieved by integrating dynamic multiscale contact and coordinated regulation through temperature-mediated switchable hydrogen bond interactions in phase-separated smart hydrogels.

The potent pro-inflammatory cytokine, interferon gamma (IFN-γ), is an enticing therapeutic target because of its accelerator role in several acute and chronic inflammatory processes. In this work, poloxamer 407 is developed as an in-situ gelling polymer for a long-acting formulation to deliver a serine protease, C5a peptidase (ScpA) from Streptococcus pyogenes. ScpA is well known for its activity against the complement factor C5a but has also recently been shown to cleave IFN-γ in vitro into inactive fragments.
