Vincristine (VCR) is a commonly used clinical anti-cancer drug, but it can also induce neurotoxicity and cause vincristine-induced neuropathic pain (VINP). The metabotropic glutamate receptor 5 (mGluR5) within spinal dorsal horn neurons regulates the transmission of pain mediated by glutamate. In this study, we investigated for the first time the role of mGluR5 in the transmission of noxious information in VINP.

X-linked Charcot-Marie-Tooth disease (CMT1X) is an inherited demyelinating neuropathy caused by loss-of-function mutations in the GJB1 gene, encoding the gap junction protein connexin32 (Cx32). Cx32 plays a critical role in Schwann cell function and myelin formation in the peripheral nervous system. We have developed a gene replacement therapeutic approach using a humanized AAVrh10 vector construct expressing GJB1 under the control of the Schwann cell-specific human myelin protein zero (MPZ) promoter.

Objectives: This research aims to better delineate how intrathecal drug delivery systems (IDDS) are incorporated into the oncology care continuum and highlight the need for further awareness of interventional options for pain management of cancer patients in the United Kingdom. The study focuses on exploring the knowledge, perspectives, and experiences of healthcare professionals regarding IDDS as a treatment option for managing chronic refractory pain in cancer patients.
Methodology: A thematic coding using inductive analysis was employed to achieve the research objectives.

Background: Perioperative neurocognitive disorders (PND) are common in elderly patients after surgery, leading to long-term cognitive decline and reduced quality of life. The mechanisms are unclear, but ferroptosis, a key cell death pathway, may be involved in the disruption of brain homeostasis during perioperative stress.
Methods: In this study, we used the SAM-P8 mouse model to simulate brain aging and observe isoflurane-induced ferroptosis.
