Leishmaniases, caused by Leishmania parasites, are widespread and pose significant health risks globally. Visceral leishmaniasis (VL) is particularly prevalent in Brazil, with high morbidity and mortality rates. Traditional treatments, such as pentavalent antimonials, have limitations due to toxicity and resistance. Therefore, exploring new compounds like lectins is crucial. Concanavalin A (ConA) has shown promise in inhibiting Leishmania growth. This study aimed to evaluate its leishmanicidal effect on L. infantum promastigotes and understand its mechanism of action. In vitro tests demonstrated inhibition of promastigote growth when treated with ConA, with IC values ranging from 3 to 5 μM over 24-72 h. This study suggests that ConA interacts with L. infantum glycans. Additionally, ConA caused damage to the membrane integrity of parasites and induced ROS production, contributing to parasite death. Scanning electron microscopy confirmed morphological alterations in treated promastigotes. ConA combined with the amphotericin B (AmB) showed synergistic effects, reducing the required dose of AmB, and potentially mitigating its toxicity. ConA demonstrated no cytotoxic effects on macrophages, instead stimulating their proliferation. These findings reinforce that lectin exhibits promising leishmanicidal activity against L. infantum promastigotes, making ConA a potential candidate for leishmaniasis treatment.

This work describes the synthesis and biological evaluation of hitherto unknown N-arylspermidine derivatives 3. Compounds 3 were efficiently prepared from cyclic amidines through a novel synthetic approach comprising alkylation with ω-halonitriles followed by reduction. The cyclic N-arylamidine directs the alkylation to the unsubstituted nitrogen and also provides the N-benzyl group present in the triamine after simultaneous reduction of the resulting quaternary salt 2 and the cyano group.

Background: Cutaneous Leishmaniases (CL), highly endemic in Africa and Mediterranean region, are caused by different Leishmania parasite species. Accurate species identification is crucial for effective diagnosis, treatment, and control of these diseases, but traditionally relies on DNA-based methods. High Resolution Melting analysis PCR (HRM PCR) provides rapid results and precise differentiation based on nucleotide variations.

Article Synopsis
  • * The researchers tested the new Cs/PF/MTF formulation's effectiveness against both the parasite stages in vitro and in vivo on naturally infected dogs, finding it significantly more potent than standard miltefosine.
  • * The new formulation was well-tolerated in dogs, with successful intravenous doses of 170 μg/kg administered daily for 28 days, indicating potential for improved treatment outcomes in CVL.

Article Synopsis
  • Leishmaniasis is a major public health concern in Iran, and current treatments like pentavalent antimonial drugs have significant drawbacks, including drug resistance and toxicity.
  • This study investigates the effectiveness of green-synthesized silver nanoparticles (Ag-NPs) derived from ginger extract in treating leishmaniasis by evaluating their toxicity on macrophage cells and their effect on leishmania promastigotes.
  • Results show that Ag-NPs significantly reduce the growth of promastigotes, with higher doses leading to increased programmed cell death, making them a promising alternative for leishmaniasis treatment.
