The development of anticancer drugs based on zinc-dependent histone deacetylase inhibitors (HDACi) has acquired great practical significance over the past decade. The most important HDACi characteristics are selectivity and strength of inhibition since they determine the mechanisms of therapeutic action. For in-cell testing of the selectivity of de novo-synthesized HDACi, Western blot analysis of the level of acetylation of bona fide protein substrates of HDACs of each class is usually used. However, the high labor intensity of this method prevents its widespread use in inhibitor screening. We developed an in-cell high-throughput screening method based on the use of three subtype-selective fluorogenic substrates of the general structure Boc-Lys(Acyl)-AMC, which in many cases makes it possible to determine the selectivity of HDACi at the class level. However, we found that the additional inhibitory activity of HDACi against metallo-β-lactamase domain-containing protein 2 (MBLAC2) leads to testing errors.

This study investigates the synthesis of ZnSnO@SiO@5-FU nanoparticles as an additive for bone fillers in dental maxillofacial reconstruction. ZnSnO nanoparticles were synthesized and coated with a SiO shell, followed by the incorporation of 5-Fluorouracil (5-FU), aimed at enhancing the therapeutic properties of classical fillers. Structural analysis using X-ray diffraction confirmed that ZnSnO was the single crystalline phase present, with its crystallinity preserved after both SiO coating and 5-FU incorporation.

Azulene-1,3-bis(semicarbazone), , and azulene-1,3-bis(thiosemicarbazone), , were synthesized by the acid-catalyzed condensation reactions of semicarbazide and thiosemicarbazide, respectively, with azulene-1,3-dicarboxaldehyde in stoichiometric amounts. Compounds and were identified by high-resolution mass spectrometry and characterized by IR, H-NMR, C-NMR, and UV-vis spectroscopic techniques. Crystal structure determination of azulene-1,3-bis(thiosemicarbazone) shows that the thiosemicarbazone units exhibit a -closed conformation, with both arms oriented in the same direction and adopting an configuration with respect to the imine linkages.
