SARS-CoV-2 evolves gradually to cause COVID-19 epidemic. One of driving forces of SARS-CoV-2 evolution might be activation of apolipoprotein B mRNA editing catalytic subunit-like protein 3 (APOBEC3) by inflammatory factors. Here, we aimed to elucidate the effect of the APOBEC3-related viral mutations on the infectivity and immune evasion of SARS-CoV-2. The APOBEC3-related C > U mutations ranked as the second most common mutation types in the SARS-CoV-2 genome. mRNA expression of (), (), and () in peripheral blood cells increased with disease severity. , a critical member of the APOBEC3 family, was significantly upregulated in both severe and moderate COVID-19 patients and positively associated with neutrophil proportion and COVID-19 severity. We identified USP18 protein, a key molecule centralizing the protein-protein interaction network of key APOBEC3 proteins. Furthermore, mRNA expression of was significantly correlated to and expression in the tissue of upper airways. Knockdown of 8 mRNA significantly decreased expression. Ectopic expression of gene increased SARS-CoV-2 infectivity. C > U mutations at S371F, S373L, and S375F significantly conferred with the immune escape of SARS-CoV-2. Thus, APOBEC3, whose expression are upregulated by inflammatory factors, might promote SARS-CoV-2 evolution and spread via upregulating USP18 level and facilitating the immune escape. A3B and USP18 might be therapeutic targets for interfering with SARS-CoV-2 evolution.

Virus-encoding RNA-dependent RNA polymerase (RdRp) is essential for genome replication and gene transcription of human coronaviruses (HCoVs), including severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). We previously identified the interaction between the catalytic subunit NSP12 of SARS-CoV-2 RdRp and the host protein CREB-regulated transcription coactivator 3 (CRTC3), a member of the CRTC family that regulates cyclic AMP response element-binding protein (CREB)-mediated transcriptional activation. Objectives: To understand the epidemic characteristics of various SARS-CoV-2 variants, we mainly focus on analyzing general epidemic profiles, viral mutation, and evolution of COVID-19 outbreaks caused by different SARS-CoV-2 variants of concern (VOCs) in China as of August 2022.
Methods: We systematically sorted out the general epidemic profiles of outbreaks caused by various SARS-CoV-2 VOCs in China, compared the differences of outbreaks caused by Delta and Omicron VOCs, and analyzed the mutational changes of subvariants between the same outbreak and different outbreaks.
Findings: By 15 August 2022, a total of 2, 33, and 124 COVID-19 outbreaks caused by Alpha, Delta, and Omicron VOCs, respectively, were reported in different regions of China.

Background: No data are available regarding the interplay and clinical manifestations of respiratory syncytial virus (RSV) and severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) coinfection in African children. We compared clinical characteristics and outcomes between RSV-only, SARS-CoV-2-only and RSV/SARS-CoV-2 coinfection lower respiratory tract infections (LRTI) in hospitalized African children.
Methods: Prospective surveillance of children (0-59 months) hospitalized with severe LRTI was undertaken between March 1, 2020, and March 31, 2023, in Johannesburg, South Africa.

In this review, we summarize ongoing efforts to develop antibodies targeting various epitopes of the S protein, focusing on their neutralization potency, breadth, and escape mechanisms. Antibodies targeting the receptor-binding site (RBS) typically exhibit high neutralizing potency but are frequently evaded by mutations in SARS-CoV-2 variants.

As pathogens spread in a population of hosts, immunity is built up, and the pool of susceptible individuals are depleted. This generates selective pressure, to which many human RNA viruses, such as influenza virus or SARS-CoV-2, respond with rapid antigenic evolution and frequent emergence of immune evasive variants. However, the host's immune systems adapt, and older immune responses wane, such that escape variants only enjoy a growth advantage for a limited time.
