This manuscript reports on the synthesis and characterization of a new polymeric copper complex ([Cu(DMAP)(μ-CO)]I) ·HO and its successful application in C-O and C-S cross coupling reactions for the synthesis of biologically important phenoxypyrimidine and arylthiopyrimidine scaffolds. In an attempt to synthesize [Cu(DMAP)I]I by adopting a procedure reported by Roy with slight modification, the authors discovered a new polymeric Cu-complex that contains μ-CO bridges. The polymeric linear structure of the complex was established using single crystal X-ray analysis. FT-IR, UV-vis and DSC studies were also performed on the polymeric complex. This novel polymeric Cu-complex was found to efficiently catalyse C-O/C-S cross coupling reactions between chloropyrimidines and phenols/thiophenols in an aqueous medium within a short reaction time, delivering their corresponding phenoxypyrimidines and arylthiopyrimidines. Using this protocol, 22 phenoxypyrimidines and 6 arylthiopyrimidines were successfully synthesized. The synthesized novel compounds were well characterized using H and C NMR spectroscopy and HRMS analysis and were screened for their drug-likeness properties using the SwissADME webtool.

The synthetical methodology for the [Cu(dmp)] (dmp = 2,9-dimethyl-1,10-phenanthroline; neocuproine) complexes has been systematically investigated by using various copper precursors, including CuCl, Cu(NO), and Cu(ClO). After an anion exchange to trifluoromethanesulfonimide (TFSI), the tetra-coordinated Cu(dmp)(TFSI)-Cu(ClO) (7.43%) outperformed the penta-coordinated Cu(dmp)(TFSI)(NO)-Cu(NO) (4.

Article Synopsis
  • Melanoma is a serious health concern, and the study examines the combination of disulfiram (DS) and copper (Cu) for a safer and more effective cancer treatment via encapsulation in a polyvinyl alcohol (PVA) scaffold.
  • The DS-Cu@PVA scaffold was created using an electrospinning technique and analyzed for its properties, showing successful encapsulation and minimal changes to PVA's structure.
  • Results indicated that the scaffold is biocompatible and effective at inducing apoptosis in melanoma cells while maintaining low toxicity to normal skin cells, suggesting a promising approach for melanoma treatment.

Doxorubicin (DOX) is a chemotherapeutic drug used in osteosarcoma treatments, usually administrated in very high dosages. This study proposes novel DOX microcarriers based on chitosan (CHT) physically crosslinked with copper(II) ions that will act synergically to inhibit tumor growth at lower drug dosage without affecting the healthy cells. Spherical CHT-Cu microparticles with a smooth surface and an average size of 30.

In catalysis, linear free energy relationships (LFERs) are commonly used to identify reaction descriptors that enable the prediction of outcomes and the design of more effective catalysts. Herein, LFERs are established for the reductive cleavage of the C(sp)-X bond in alkyl halides (RX) by Cu complexes. This reaction represents the activation step in atom transfer radical polymerization and atom transfer radical addition/cyclization.
