Background: Rheumatoid arthritis (RA) is a chronic autoimmune disease with abnormal differentiation of follicular helper T (Tfh) cells, Total alkaloids of Linn. (Leguminosae) (TASA) have potential effects on collagen-induced arthritis (CIA) mice, while the mechanism needs further elucidation. The purpose of this study is to explore the regulation of TASA on rheumatoid arthritis and related mechanism.

Methods: The proportion of Tfh and B lymphocytes in peripheral blood lymphocytes of RA patients was examined by flow cytometry. We constructed the collagen induced arthritis DBA/1J mice model. Between days 15 and 45 following the first immunization, the mice were treated intraperitoneally with saline, TASA (100, 50, and 25 mg/kg), and dexamethasone (DXM) for 30 days. Molecular biological techniques such as FCM, PCR, ELISA, and Western-blotting were used to examine Tfh cells and associated signal pathways.

Results: Our results indicated that the follicular helper T cells and B lymphocytes in rheumatoid arthritis patients were significantly increased compared with the healthy control. The percentage of Tfh cells are correlated with RA related inflammatory factors. Total alkaloids of Linn. could significantly attenuate joint swelling. Meanwhile, it reduced the frequencies of spleen Tfh, B lymphocytes and the expression of TLR2, TLR9, p-NF-κBp65, CXCR5, Bcl-6, ICOS of ankle joints in CIA mice.

Conclusion: Total alkaloids of Linn. may down-regulate the frequency and function of Tfh cells and inhibit GCB cells via TLRs/NF-κB signal pathway to relieve the immune-pathological progression of CIA mice.

Purpose: To create a computer-aided prediction (CAP) system to predict Wilms tumor (WT) responsiveness to preoperative chemotherapy (PC) using pre-therapy contrast-enhanced computed tomography (CECT).

Materials And Methods: A single-center database was reviewed for children <18 years diagnosed with WT and received PC between 2001 and 2021. Patients were excluded if pre- and post-PC CECT were not retrievable.

Objectives:  Hochst, belonging to the  family, is known for its ethnomedicinal values, particularly in treating various diseases in Africa. This study aimed to investigate the antibacterial, antioxidant, and cytotoxic properties of stem bark extracts, with a specific focus on assessing their bioactive potential and identifying active compounds via TLC bioautography.

Methods: The antimicrobial activity was assessed using the agar well diffusion method against selected clinical strains.

Since antiquity, alkaloid natural products have served as medicinal ingredients that still contribute as an inspiration for the development of novel therapeutics. For the synthetic chemist, much of the importance of natural products lies in their acting as a forcing-function for the invention of new synthetic strategies and tactics for molecular assembly. With this rich history in mind, it remains an important goal for chemists to build nitrogenous structures with greater efficiency, abiding by economies of synthesis.

Department of Neurology, Graduate School of Medicine, Chiba University, Chiba, Japan.

Background: Dopamine transporter (I-FP-CIT) single-photon emission tomography (SPECT) and I-meta-iodobenzylguanidine (I-MIBG) image play roles as indicative biomarkers in diagnosing patients with dementia with Lewy bodies (DLB). Brain- and body-first subtypes of DLB were hypothesized implying that subset of DLB may have normal I-FP-CIT or I-MIBG results, respectively. The purpose of this study was to explore the diagnostic sensitivity of two combination imaging modalities (I-FP-CIT SPECT and I-MIBG image) in patients with DLB and examine the clinical difference between brain- and body-first subtype.

Background: We aimed to investigate the association between maternal caffeine intake during pregnancy and asthma in children by 10 years of age.

Methods: We considered 5585 mother-child pairs enrolled in a population-based birth cohort. Consumption of regular and decaffeinated coffee, black and green tea, and cola beverages before and during pregnancy was obtained through face-to-face interviews within 72 h after giving birth, and total caffeine intake (mg/day) was estimated.
