Activity-based detection of hydrogen sulfide in live cells can expand our understanding of its reactivity and complex physiological effects. We have discovered a highly efficient method for fluorescent probe activation, which is driven by HS-triggered 1,6-elimination of an α-CF-benzyl to release resorufin. In detecting intracellular HS, 4-azido-(α-CF)-benzyl resorufin offers significantly faster signal generation and improved sensitivity compared to 4-azidobenzyl resorufin. Computed free energy profiles for the 1,6-elimination process support the hypothesis that a benzylic CF group can reduce the activation energy barrier toward probe activation. This novel probe design allows for near-real-time detection of HS in HeLa cells under stimulation conditions.

Peptide stapling is an effective strategy to stabilise α-helical peptides, enhancing their bioactive conformation and improving physiochemical properties. In this study, we apply our novel diyne-girder stapling approach to the MDM2/MDMX α-helical binding region of the p53 transactivation domain. By incorporation of an unnatural amino acid to create an optimal , + 7 bridge length, we developed a highly α-helical stapled peptide, 4, confirmed circular dichroism.

Tidal interactions play a key role in the dynamics and evolution of icy worlds. The intense tectonic activity of Europa and the eruption activity on Enceladus are clear examples of the manifestation of tidal deformation and associated dissipation. While tidal heating has long been recognized as a major driver in the activity of these icy worlds, the mechanism controlling how tidal forces deform the different internal layers and produce heat by tidal friction still remains poorly constrained.
