The global prevalence of fungal infections is alarming in both the pre- and post- COVID period. Due to a limited number of antifungal drugs, there are hurdles in treatment strategies for fungal infections due to toxic potential, drug interactions, and the development of fungal resistance. All the antifungal targets (existing and newer) and pipeline molecules showing promise against these targets are reviewed.

Autocrine regulation of tumor cell repopulation by Hsp70-HMGB1 alarmin complex.

J Exp Clin Cancer Res

October 2023

Department of Molecular and Cellular Interaction, Institute of Cytology of Russian Academy of Sciences, Tikhoretsky prospect, 4, St. Petersburg, 194064, Russia.

Background: Cancer recurrence is regulated by a variety of factors, among which is the material of dying tumor cells; it is suggested that remaining after anti-cancer therapy tumor cells receive a signal from proteins called damage-associated molecular patterns (DAMPs), one of which is heat shock protein 70 (Hsp70).

Methods: Two models of tumor repopulation were employed, based on minimal population of cancer cells and application of conditioned medium (CM). To deplete the CMs of Hsp70 affinity chromatography on ATP-agarose and immunoprecipitation were used.

Asymptotic theory of not completely integrable soliton equations.

Chaos

September 2023

Institute of Spectroscopy, Russian Academy of Sciences, Troitsk, Moscow 108840, Russia.

We develop the theory of transformation of intensive initial nonlinear wave pulses to trains of solitons emerging at asymptotically large time of evolution. Our approach is based on the theory of dispersive shock waves in which the number of nonlinear oscillations in the shock becomes the number of solitons at the asymptotic state. We show that this number of oscillations, which is proportional to the classical action of particles associated with the small-amplitude edges of shocks, is preserved by the dispersionless flow.

Quantitatively Accounting for Protein Reorganization in Computer-Aided Drug Design.

J Chem Theory Comput

June 2023

Schrodinger Inc., 1540 Broadway, New York, New York 10036, United States.

Structure-based drug design frequently operates under the assumption that a single holo structure is relevant. However, a large number of crystallographic examples clearly show that multiple conformations are possible. In those cases, the protein reorganization free energy must be known to accurately predict binding free energies for ligands.
