AI Article Synopsis

  • The study presents two new synthetic riboswitches designed to respond to specific molecules, ASP2905 and theophylline.
  • These riboswitches were integrated into a cell-free protein synthesis (CFPS) system that was encapsulated in liposomes.
  • External stimulation using the corresponding ligands allowed for precise control over gene expression regulated by the riboswitches.

Article Abstract

Here we report two novel synthetic riboswitches that respond to ASP2905 and theophylline and function in reconstituted cell-free protein synthesis (CFPS) system. We encapsulated the CFPS system as well as DNA-templated encoding reporter genes regulated by these orthogonal riboswitches inside liposomes, and achieved switchable and orthogonal control over gene expression by external stimulation with the cognate ligands.

Chiral organic-inorganic hybrid perovskites offer a promising platform for developing non-linear chiro-optical applications and chiral-induced spin selectivity. Here, we show that achiral hybrid perovskites that have highly ordered ferroelectric domains with orthogonal polarization exhibit planar chirality, as manifested by second harmonic generation with strong circular dichroism. Interestingly, the handedness of the second harmonic generation circular dichroism response can be alternatingly switched between orthogonally polarized domains and domain walls.

Droplet manipulation technologies with surface acoustic waves attract significant attention for applications in fluid handling and bioanalysis. However, existing technologies face challenges in automation, precision, and functional integration, limiting broader applications. In this work, a highly integrated droplet-sensing acoustofluidic tweezer is developed, incorporating orthogonally arranged slanted finger interdigital transducers and a custom-designed control and detection circuit system.

Background: De-novo-designed synthetic transcriptional regulators have great potential as the genetic parts for constructing complex multilayered gene circuits. The design flexibility afforded by advanced nucleic acid sequence design tools vastly expands the repertoire of regulatory elements for circuit design. In principle, the design space of synthetic regulators should allow for the construction of regulatory circuits of arbitrary complexity; still, the orthogonality and robustness of such components have not been fully elucidated, thereby limiting the depth and width of synthetic circuits.

Edge detection is a fundamental operation for feature extraction in image processing. The all-optical method has aroused growing interest owing to its ultra-fast speed, low energy consumption and parallel computation. However, current optical edge detection methods are generally limited to static devices and fixed functionality.

Based on the unique insulator-metal phase transition property of vanadium dioxide (VO), we propose an integrated metasurface with a switchable mechanism between ultra-broadband absorption and polarization conversion, operating in the terahertz (THz) frequency range. The designed metasurface device is constructed using a stacked structure composed of VO quadruple rings, a dielectric layer, copper stripes, VO film, a dielectric layer, and a copper reflection layer. Our numerical simulations demonstrate that our proposed design, at high temperatures (above 358 K), exhibits an ultra-broadband absorption ranging from 4.
