Background: The microvasculature of the central nervous system (CNS), which delivers oxygen and nutrients and forms a critical barrier protecting the CNS, is deleteriously affected by both Alzheimer's Disease (AD) and Type 2 Diabetes (T2D). Previous studies have shown pericyte dropout and vessel constriction in brain capillaries in AD, while other studies have shown pericyte bridging and dropout in retinal capillaries in T2D. T2D patients have increased risk of AD, suggesting potentially related microvascular pathological mechanisms.

Background: Amyotrophic lateral sclerosis (ALS) with only motor impairment (ALS-pure motor) and the behavioral variant of frontotemporal dementia (bvFTD) are hypothesized to be the extreme ends of the ALS-bvFTD spectrum. This spectrum also encompasses ALS patients with mild to severe cognitive impairment (ALSci) and/or behavioral impairment (ALSbi), including ALS with concomitant bvFTD. In a previous study, using magnetoencephalography (MEG), in early symptomatic ALS patients we showed resting-state functional connectivity changes in frontal, limbic and subcortical regions that overlap considerably with bvFTD.

Work has progressed from clinical, to neuroimaging to biomedical devices to neuropathological data, and now brain and blood omic data.
Method: The AMP-AD consortium generated and/or annotated genomic, epigenomic, transcriptomic, proteomic, and metabolomic data from brain and/or blood from thousands of study participants and patients across the 8 teams.

Background: When performing a picture description task, healthy individuals tend to look only briefly at a target before beginning its description, after which they move promptly onto the next target. This sequence may be disrupted in those with cognitive impairment. Just as cognitively impaired individuals produce greater numbers of disfluencies and pauses, those with mild cognitive impairment (MCI) may delay speech production by extending their gaze behavior towards a target before beginning its description.
