Ophiobolin A (OPA) is a sesterterpenoid fungal natural product with broad anticancer activity. While OPA possesses multiple electrophilic moieties that can covalently react with nucleophilic amino acids on proteins, the proteome-wide targets and mechanism of OPA remain poorly understood in many contexts. In this study, we used covalent chemoproteomic platforms to map the proteome-wide reactivity of the OPA in a highly sensitive lung cancer cell line. Among several proteins that OPA engaged, we focused on two targets: lysine-72 of cytochrome c oxidase subunit 5A (COX5A) and cysteine-53 of mitochondrial hypoxia induced gene 1 domain family member 2A (HIGD2A). These two subunit proteins are part of complex IV (cytochrome C oxidase) within the electron transport chain and contributed significantly to the antiproliferative activity of OPA. OPA activated mitochondrial respiration in a COX5A- and HIGD2A-dependent manner, leading to an initial spike in mitochondrial ATP and heightened mitochondrial oxidative stress. OPA compromised mitochondrial membrane potential, ultimately leading to ATP depletion. We have used chemoproteomic strategies to discover a unique anticancer mechanism of OPA through activation of complex IV leading to compromised mitochondrial energetics and rapid cell death.

The study of scene perception is crucial to the understanding of how one interprets and interacts with their environment, and how the environment impacts various cognitive functions. The literature so far has mainly focused on the impact of low-level and categorical properties of scenes and how they are represented in the scene-selective regions in the brain, PPA, RSC, and OPA. However, higher-level scene perception and the impact of behavioral goals is a developing research area.

Silver nanowire (Ag NW)/gold nanosphere (Au NS) binary plasma films were prepared using plasma coupling between Ag NWs and Au NSs. The plasma films formed by combining these two noble metals showed better sensitivity for SERS detection with a minimum detection concentration of 10 M for R6G compared to pure Ag NWs or Au NSs. After rational optimisation of the substrate preparation process, the substrate showed good homogeneity, reproducibility and stability.

In this paper, a new carbon dot (R1-CDs) was prepared by one-pot hydrothermal method by using 1,8-diaminonaphthalene and o-phthalic acid (o-PA) as precursors. Due to the high purity of R1-CDs, NMR analysis was performed to identify the types of H and C atoms in their graphene sheets. From our research findings, three important information was disclosed such as (1) five types H atoms are presented in R1-CDs; (2) 18 kinds of C atoms in the graphene sheets are observed, and 8 kinds of them are quaternary atoms, and 10 kinds of carbon atoms as tertiary one; (3) functional groups of -COOH and -NH2 from precursors cannot be inherited into the edges or defect sites of graphene sheet.

Herein, UiO-66 was selected as the immobilization carrier of Rhizomucor miehei lipase (RML). After etching and hydrophobic modification, the functionalized UIO-66 (H-UIO-66-OPA) was utilized for RML immobilization and the obtained RML@H-UIO-66-OPA showed about 70 % relative activity after incubation at 60 °C, which was much better than RML (20 %). RML@H-UIO-66-OPA was used in the synthesis of 1,3-dioleic acid glycerol (1,3-DAG) and the effects of reaction conditions (temperature, enzyme addition, substrate molar ratio, and time) on 1,3-DAG yield were investigated.
