Autotaxin, encoded by the gene, produces lysophosphatidic acid (LPA), which exerts numerous biological functions via its cognate receptors. null mutant mice die by embryonic day 9.5 owing to aberrant vascular development in the yolk sac, preventing analysis after that period. In this study, we found that heterozygous mice in the DBA/2 genetic background showed an eye-open-at-birth phenotype at high frequency, caused by failure of eyelid closure during the embryonic stage. Notably, wildtype pups from the heterozygous dam showed the phenotype, although at lower frequency, suggesting that maternal LPA affects the embryonic development.

Various techniques using fluorescent reporter probes have been developed, such as GFP transgenic mouse lines that are used to detect spatial-temporal expression levels of genes. Although GFP expression is largely considered non-toxic, recent reports have indicated that under certain conditions GFP can display cellular toxicity. We hereby report the nuclear toxicity of H2B-GFP using a K14 specific Tet-on reporter mouse system.

Crosstalk of MAP3K1 and EGFR signaling mediates gene-environment interactions that block developmental tissue closure.

Aberrant regulation of signal transduction pathways can adversely derail biological processes for tissue development. One such process is the embryonic eyelid closure that is dependent on the mitogen-activated protein kinase kinase kinase 1 (MAP3K1). Map3k1 KO in mice results in defective eyelid closure and an autosomal recessive eye-open at birth phenotype.

An L314Q mutation in Map3k1 gene results in failure of eyelid fusion in the N-ethyl-N-nitrosourea-induced mutant line.

Article Synopsis
  • This study introduces a new mouse model that exhibits corneal opacity and an "eye open at birth" (EOB) condition, caused by a mutation induced by N-ethyl-N-nitrosourea.
  • The abnormal traits in the corneal epithelial cells of these mutant mice are linked to a specific genetic mutation in the MAP3K1 gene, identified as a T941A substitution leading to the L314Q amino acid change.
  • Findings suggest that the EOB phenotype is connected to lower levels of certain proteins (p-JNK and c-Jun) that are essential for normal epithelial cell function and migration.

Background: The timing, location, and level of gene expression are crucial for normal organ development, because morphogenesis requires strict genetic control. MicroRNAs (miRNAs) are noncoding small single-stranded RNAs that play a critical role in regulating gene expression level. Although miRNAs are known to be involved in many biological events, the role of miRNAs in organogenesis is not fully understood.
