Polyphyllin VII is a biologically active herbal monomer extracted from the traditional Chinese herbal medicine Chonglou. Many studies have demonstrated the anticancer activity of polyphyllin VII against various types of cancers, such as colon, liver, and lung cancer, but its effect on breast cancer has not been elucidated. In this study, we demonstrate that polyphyllin VII inhibited proliferation, increased production of intracellular reactive oxygen species, and decreased mitochondrial membrane potential in breast cancer cells. Notably, polyphyllin VII also induced apoptosis via the mitochondrial pathway. Transcriptome sequencing was used to analyze the targets of PPVII in regulating breast cancer cells. Mechanistic studies showed that polyphyllin VII downregulated Son of Sevenless1 (SOS1) and inhibited the MAPK/ERK pathway. Furthermore, PPVII exerted strong antitumor effects in nude mice injected with breast cancer cells. Our results suggest that PPVII may promote apoptosis through regulating the SOS1/MAPK/ERK pathway, making it a possible candidate target for the treatment of breast cancer.

Article Synopsis
  • Endometrial carcinoma (EC) often resists hormone therapies like medroxyprogesterone acetate (MPA), necessitating new treatment approaches.
  • This study examines how polyphyllin VII (PPVII) can improve MPA effectiveness by regulating microRNA (miR)-33a-5p in an MPA-resistant Ishikawa cell line.
  • Results showed that combining PPVII with MPA significantly reduced cell viability and increased apoptosis, while PPVII enhanced miR-33a-5p levels, suggesting a potential therapeutic strategy for overcoming hormone resistance in EC.

Article Synopsis
  • Smith var. (Franch.) Hara is a medicinal plant with beneficial components called parissaponins, known for their anti-inflammatory and anti-tumor effects, prompting research on enhancing these compounds through UV treatments.
  • * In a study, UV treatments (UVA, UVB, UVC) resulted in a significant increase in PolyPhyllin VII (PPVII) content by 32.43% with UVC after 4 hours, while other treatments decreased relevant saponin levels.
  • * The study found that UV exposure activated several enzymes involved in saponin synthesis and affected various physiological parameters, providing insights into using UV light for improving medicinal properties in plants, which is valuable for future production techniques.*

The need for novel anti-thyroid cancer (TC) medications is urgent due to the rising incidence and metastatic rates of malignant TC. In this study, we investigated the effect of Polyphyllin VII (PPVII) to TC cells, and explored their potential mechanism. B-CPAP and TPC-1 cells, were used to analyze the antitumor activity of PPVII by quantifying cell growth and metastasis as well as to study the effect on epithelial mesenchymal transition (EMT).
