The immobilisation of artificial metalloenzymes (ArMs) holds promise for the implementation of new biocatalytic reactions. We present the synthesis of cross-linked artificial metalloenzyme aggregates (CLArMAs) with excellent recyclability, as an alternative to carrier-based immobilisation strategies. Furthermore, iron-siderophore supramolecular anchoring facilitates redox-triggered cofactor release, enabling CLArMAs to be recharged with alternative cofactors for diverse selectivity.

Cancer stem cells (CSCs) have been implicated as critical mediators in the progression, chemoresistance and metastatic capabilities of diverse malignancies, including osteosarcoma (OS). The authors have succeeded in generating CSC‑like cells (MG‑OKS) from the OS cell line MG‑63 by transducing defined factors. A significant increase in small proline‑rich protein 1A (SPRR1A) expression, a cross‑linked envelope protein in keratinocytes, was observed in MG‑OKS cells.

3D Printed GelMA/CHIMA Cross-Linked Network Hydrogel for Angiogenesis.

Biotechnol Bioeng

December 2024

Shanxi Key Laboratory of Micro Nano Sensors & Artificial Intelligence Perception, College of Integrated Circuits, Taiyuan University of Technology, Taiyuan, China.

Vascularization is a key issue facing the construction of functional three-dimensional (3D) tissues, which is critical for the long-term survival and stability of tissue construct transplantation. In this study, a photocurable hydrogel material carboxymethyl chitosan (CHIMA) was successfully prepared and integrated with methacryloyl gelatin (GelMA) to construct the bioink GelMA/CHIMA, which was subsequently used 3D printing technology to prepared a bioactive scaffold with angiogenesis-inducing functionality. The results showed that the cross-linked GelMA/CHIMA bioink had a porous structure that supported cell growth and metabolism.

Polypeptide-Folded Artificial Ferroprotein Promotes Ferroptosis in Multiple Tumor Cells.

Biomacromolecules

December 2024

Beijing Area Major Laboratory of Peptide and Small Molecular Drugs, School of Pharmaceutical Sciences, Capital Medical University, Beijing 100069, China.

Although the current nanozymes, such as FeO nanoparticles, exhibit biocatalytic activities, they dramatically differ from natural enzymes, lacking a degradable organic framework and an intrinsically flexible structure. Single-chain folding of a synthetic polypeptide by metal coordination can mimic metalloproteins more similarly. A triblock PEG-polypeptide copolymer, poly(ethylene glycol)--poly(but-3-yn-1-yl glutamate)--poly(-butyl glutamate) [EG(Glu-yne)(Glu-tBu)], was synthesized by NCA polymerization.

High-risk neuroblastoma (HR-NB) patient treatment is currently insufficient and challenging due to its high clinical, morphological, and genetic heterogeneity as well as the scarcity of available samples for research. We used a gelatin- and silk fibroin-based hydrogel system with cross-linked vitronectin (VN) as an artificial biomimetic three-dimensional (3D) environment to mirror aggressive neuroblastoma (NB) tumors and tested long-term cell response to Cilengitide (CLG). Based on our previous studies and others using the integrin inhibitor CLG as a potential mechanotherapy drug, we show that CLG caused cell detachment in monolayer cultures of -amplified SK-N-BE (2) and -mutated SH-SY5Y human neuroblastoma cell lines.

Objective: This study aimed to assess the occlusal contact area (OCA), occlusal contact number (OCN), bite force, and artificial tooth wear in complete dentures with lingualized and bilateral balanced occlusion.

Methods: Edentulous participants who fulfilled the inclusion criteria were divided into the three groups as follows: group I, dentures with lingualized occlusion using interpenetrating polymer network artificial teeth; group II, dentures with lingualized occlusion using double cross-linked artificial teeth; and group III, dentures with bilateral balanced occlusion using anatomical micro-hybrid resin teeth. The silicone techniques were used to assess the OCA and OCN of the mandibular dentures and the Dental Prescale II was used to evaluate the bite force at the 2-week, 3-month, and 12-month follow-up visits.
