Many proteins self-assemble into dimers and higher-order oligomers. Therefore, the goal of this protocol is to characterize the conformational states of an endogenous protein of interest. Here, we present a protocol for assessing protein self-assembly in cell lysates using chemical cross-linking. We describe steps for chemical cross-linking with recombinant proteins as well as steps for cell culture and cell lysate preparation, chemical cross-linking, SDS-PAGE, and western blotting for the detection of endogenous proteins. Severe traumatic bleeding and chronic diabetic wounds require rapid hemostasis and multifunctional dressings, which remain particularly challenging, especially for non-compressible trauma and irregular wounds with dysregulated microenvironments. Chitosan (CS) can be easily cross-linked with genipin to form GpCS hydrogels. However, developing injectable GpCS hydrogels for biomedical applications faces challenges, particularly in enhancing rapid gel formation and optimizing physical properties.

Self-assembly in supramolecular chemistry is crucial for nanostructure creation but faces challenges like slow speeds and lack of reversibility. In this study, a novel comb-like polymer poly(amide sulfide) (PAS) based on thiolactone chemistry is reported, which rapidly self-assemble into stable nanofibers, offering excellent robustness and reversibility in the self-assembled structure. The PAS backbone contains pairs of amide bonds, each linked to an alkyl side chain in a controlled 2:1 ratio.
