Multi-enzyme complexes designed based on scaffold proteins are a current topic in molecular enzyme engineering. They have been gradually applied to increase the production of enzyme cascades, thereby achieving effective biosynthetic pathways. This paper reviews the recent progress in the design strategy and application of multi-enzyme complexes. First, the metabolic channels in the multi-enzyme complex have been introduced, and the construction strategies of the multi-enzyme complex emerging in recent years have been summarized. Then, the discovered enzyme cascades related to scaffold proteins are discussed, emphasizing on the influence of the linker on the fusion enzyme (fusion protein) and its possible mechanism. This review is expected to provide a more theoretical basis for the modification of multi-enzyme complexes and broaden their applications in synthetic biology.

The multi-enzyme pyruvate dehydrogenase complex (PDHc) links glycolysis to the citric acid cycle and plays vital roles in metabolism, energy production, and cellular signaling. Although all components have been individually characterized, the intact PDHc structure remains unclear, hampering our understanding of its composition and dynamical catalytic mechanisms. Here, we report the in-situ architecture of intact mammalian PDHc by cryo-electron tomography.

Objectives: To investigate the impact of different pretreatment strategies and cleaning methods on the cleaning efficacy of electrosurgical instruments, aiming to propose an optimal protocol.
Methods: A total of 573 electrosurgical instruments were collected from three large-scale hospitals within two hours post-use from December 2023 to July 2024. The instruments were categorized into six group: medical alkaline or multi-enzyme cleaner, or instant foam-type multi-enzyme humectant, followed by either manual cleaning or vacuum boiling washer.

Specific cellular microenvironment, multi-enzyme complex and expensive essential cofactor make the biological manufacturing of plant chloroplast natural products (PCNPs) extremely challenging. The above difficulties have hampered the biosynthesis of capsanthin and capsorubin in the past 30 years. Here, we take capsanthin and capsorubin as examples to design an innovative microbial factory to promote the heterologous synthesis of PCPNs.
