The giant (2-3 × 10 m long) internodal cells of the aquatic plant, , exhibit a rapid (>100 × 10 m s) cyclic cytoplasmic streaming which stops in response to mechanical stimuli. Since the streaming - and the stopping of streaming upon stimulation - is easily visible with a stereomicroscope, these single cells are ideal tools to investigate mechanosensing in plant cells, as well as the potential for these cells to be anesthetized. We found that dropping a steel ball (0.88 × 10 kg, 6 × 10 m in diameter) through a 4.6 cm long tube (delivering ca. 4 × 10 J) reliably induced mechanically-stimulated cessation of cytoplasmic streaming. To determine whether mechanically-induced cessation of cytoplasmic streaming in was sensitive to anesthesia, we treated internodal cells to volatilized chloroform in a 9.8 × 10 m chamber for 2 minutes. We found that low doses (15,000-25,000 ppm) of chloroform did not always anesthetize cells, whereas large doses (46,000 and higher) proved lethal. However, 31,000 ppm chloroform completely, and reversibly, anesthetized these cells in that they did not stop cytoplasmic streaming upon mechanostimulation, but after 24 hours the cells recovered their sensitivity to mechanostimulation. We believe this single-cell model will prove useful for elucidating the still obscure mode of action of volatile anesthetics.

Pre- and post-synaptic events are regulated by liquid-liquid phase separation and this phenomenon requires multiple electrical forces. Both axonal transport and the organization of postsynaptic excitatory and inhibitory receptors are regulated by LLPS, with its mandatory electrical drivers ultimately determining our cognitive health and capacity.

Amyotrophic lateral sclerosis (ALS), commonly known as motor neuron disease, is a neurodegenerative disorder characterized by the progressive degeneration of both upper and lower motor neurons. This pathological process results in muscle weakness and can culminate in paralysis. To date, the precise etiology of ALS remains unclear.

Mutations in the microtubule-binding motor protein kinesin 5 A (KIF5A) are implicated in several adult-onset motor neuron diseases, including Amyotrophic Lateral Sclerosis, Spastic Paraplegia Type 10 and Charcot-Marie-Tooth Disease Type 2. While KIF5 family members transport a variety of cargos along axons, the specific cargos affected by KIF5A mutations remain poorly understood. Here, we generated KIF5Anull mutant human motor neurons and analyzed the impact on axonal transport and motor neuron outgrowth and regeneration in vitro.

Visualization and Analyses of Cytoplasmic Streaming in C. elegans Zygotes.

Cytoplasmic streaming is the bulk flow of cytoplasm observed, not only in plants but also in animal oocytes and embryos. The flow of viscous fluid within the cytoplasm generates forces that re-arrange intracellular organelles, such as mitotic spindles and nuclei, to regulate cell growth, migration, and polarity. Cytoplasmic streaming is established by motor proteins and the viscoelastic cytoskeleton, including the actin filaments and microtubules.
