This study introduces a one-pot isothermal amplification assay for ultrasensitive analysis of terminal deoxynucleotidyl transferase (TdT) activity. The system realizes recycled activation of CRISPR/Cas12a, enabling exceptional signal amplification. This approach maximizes the simplicity of the detection method, offering a promising avenue for molecular disease diagnosis.

A series of polyurethanes (PU-GT) were prepared using polyglycolide-block-polytetrahydrofuran-block-polyglycolide (PGA-PTHF-PGA), polytetrahydrofuran homopolymer (PTHF), glycerol, and hexamethylene diisocyanate (HDI) by a one-pot synthesis method. The non-isothermal crystallization and subsequent heating curves showed that the PTHF component in these polyurethanes could crystallize in a temperature range of -11.5~2.

The detection of heavy metal ions, particularly lead (Pb²⁺), in environmental samples is crucial for public health and safety. Current nucleic acid signal amplification methods for Pb²⁺ detection often rely on biological enzymes and are limited in applicability due to high costs, prolonged detection times, and nonspecific adsorption.
