Point-of-care testing (POCT) plays an increasingly important role in biomedical research and health care. Quantitative phase microscopes (QPMs) with good contrast, no invasion, no labeling, high speed and automation could be effectively applied for POCT. However, most QPMs are fixed on the optical platform with bulky size, lack of timeliness, which remained challenging in POCT solutions. In this paper, we proposed a plug-and-play QPM with multimode imaging based on the quantitative differential phase contrast (qDPC) method. The system employs a programmable LED array as the light source and uses the GPU to accelerate the calculation, which can realize multi-contrast imaging with six modes. Accurate phase measurement and real-time phase imaging are implemented by the proposed qDPC algorithms for quantitative phase targets and biomedical samples. A 3D electric control platform is designed for mechanical control of field of view and focusing without manual operations. The experimental results verify the robustness and high performance of the setup. Even a rookie could finish the POCT scheme for biomedical applications at the scene using the QPM with a compact size of 140 × 165 × 250 mm.

Transcription Factor EB (TFEB) controls lysosomal biogenesis and autophagy in response to nutritional status and other stress factors. Although its regulation by nuclear translocation is known to involve a complex network of well-studied regulatory processes, the precise contribution of each of these mechanisms is unclear. View Article and Find Full Text PDFPhysicians evaluate a patient's respiratory health during a physical examination by visual assessment of the work of breathing (WoB) to determine respiratory stability, and by detecting abnormal lung sounds via lung auscultation using a stethoscope to identify common pathological lung diseases, such as chronic obstructive pulmonary disease (COPD) and pneumonia. Since these assessment methods are subjective, a low-profile device used for an accurate and quantitative monitoring approach could provide valuable preemptive insights into respiratory health, proving to be clinically beneficial. To achieve this goal, we have developed a miniature patch consisting of a sensitive wideband multi-axis seismometer that can be placed on the anatomical areas of a patient's lungs to enable an effective quantification of a patient's WoB and lung sounds.

β-3 adrenoceptor (AR) counteracts the β-1 and β-2 ARs and rescues the effects of excessive catecholamines. To test the hypothesis that a β-3 AR agonist (mirabegron) can reverse the effects of isoproterenol (ISO) on ventricular fibrillation (VF), we performed optical mapping studies in six male and six female Langendorff perfused rabbit hearts at baseline and after sequential administration of ISO (100 nm), mirabegron (1000 nm), apamin (100 nm) and washout (Study I). An additional six male and six female hearts were studied with mirabegron doses ranging between 250 and 1000 nm without ISO (Study II).
