Due to their ease of preparation, stability, and diverse reactivity, -hydroxyphthalimide (NHPI) esters have found many applications as radical precursors. Mechanistically, NHPI esters undergo a reductive decarboxylative fragmentation to provide a substrate radical capable of engaging in diverse transformations. Their reduction via single-electron transfer (SET) can occur under thermal, photochemical, or electrochemical conditions and can be influenced by a number of factors, including the nature of the electron donor, the use of Brønsted and Lewis acids, and the possibility of forming charge-transfer complexes. Such versatility creates many opportunities to influence the reaction conditions, providing a number of parameters with which to control reactivity. In this perspective, we provide an overview of the different mechanisms for radical reactions involving NHPI esters, with an emphasis on recent applications in radical additions, cyclizations and decarboxylative cross-coupling reactions. Within these reaction classes, we discuss the utility of the NHPI esters, with an eye towards their continued development in complexity-generating transformations.

In this work, we used experiments and density functional theory calculations to investigate the mechanism and driving forces of the reductive fragmentation of NHPI esters. Mechanistic studies suggest that the fragmentation behavior of the NHPI ester is influenced not only by the electronic nature of the substituent group but also by the stability of the radical intermediate. To further investigate this transformation, we next examined the aminoalkoxycarbonylation of alkenes using alkyl -phthalimidoyl oxalates.

Heterocoupling Two Similar Benzyl Radicals by Dual Photoredox/Cobalt Catalysis.

Transition-metal-regulated radical cross coupling enables the selective bonding of two distinct transient radicals, whereas the catalytic method for sorting two almost identical transient radicals, especially similar benzyl radicals, is still rare. Herein, we show that leveraging dual photoredox/cobalt catalysis can selectively couple two similar benzyl radicals. Using easily accessible methylarenes and phenylacetates (benzyl N-hydroxyphthalimide (NHPI) esters) as benzyl radical sources, a range of unsymmetrical 1,2-diarylethane classes via the 1°-1°, 1°-2°, 1°-3°, 2°-2°, 2°-3° and 3°-3° couplings were obtained with broad functional group tolerance.

A practical nickel-catalyzed decarboxylative cross-coupling of -hydroxyphthalimide esters with 1,4-diiodobenzene analogues has been developed. Under mild reaction conditions, a series of structurally interesting 1,4-dialkylbenzene analogues have been accessed with good functional group tolerance, which are versatile precursors for organic synthesis and a kind of important unit for bioactive molecules, polymers, and other materials. The synthetic application of this methodology was demonstrated by the synthesis of 3-6-dialkylcarbazole derivatives.
