Synthesis of imidazo-fused polyheterocyclic molecular frameworks, viz. imidazo[1,2-]pyrrolo[3,4-]pyridines, imidazo[2,1-]pyrrolo[3,4-]isoquinolines, and benzo[]imidazo[1,2-]quinoline-6,11-diones, has been achieved by the ruthenium(II)-catalyzed [4 + 2] C-H/N-H annulation of 2-alkenyl/2-arylimidazoles with substituted maleimides and 1,4-naphthoquinones. The developed protocol is operationally simple, exhibits broad substrate scope with excellent functional group tolerance, and provides the desired products in moderate to good yields. The mechanistic studies suggest that the reaction involves the formation of a C-C bond through Ru-catalyzed C(sp)-H bond activation followed by intramolecular cyclization.

This study presents the development of thiol-maleimide/thiol-thiol double self-crosslinking hyaluronic acid-based (HA) hydrogels for use as dermal fillers. Hyaluronic acid with varying degrees of maleimide substitution (10%, 20%, and 30%) was synthesized and characterized, and HA hydrogels were fabricated using two molecular weights of four-arm polyethylene glycol (PEG10K/20K)-thiol as crosslinkers. The six resulting HA hydrogels demonstrated solid-like behavior with distinct physical and rheological properties.

A palladium-catalyzed annulation reaction of bay-iodobiphenyls with maleimides was developed. This approach allows for the efficient synthesis of a variety of 9,10-phenanthrenedicarboximides, particularly polyalkoxy-substituted 9,10-phenanthrenedicarboximides with potential liquid-crystalline properties. Research findings indicate that the resulting product, -hexyl-2,3,6,7-tetrakis(hexyloxy)-9,10-phenanthrenedicarboximide, exhibits promising, good liquid-crystalline properties.

A cobalt(III)-catalyzed dual C(sp)-H/C(sp)-H activation of 2-arylimidazopyridines and its annulation with N-substituted maleimides leads to polycyclic aromatic heterocycles. This sustainable oxidative annulation uses earth-abundant, less toxic, and cost-effective cobalt(III) catalyst that complement expensive 2nd and 3rd-row metals. This oxidative annulation features a broad substrate scope with very good functional group tolerance.

Copper radioisotopes can be used for imaging as well as for therapy and, thus, can form ideal theranostic pairs. The Cu(II) complexes of cross-bridged cyclam (cb-cyclam) derivatives are considered to be highly stable . However, the complexes are mostly formed under harsh conditions not compatible with sensitive biomolecules.

Article Synopsis
  • - Single-molecule photobleaching analysis allows researchers to study how membrane proteins form oligomers by attaching a fluorophore to individual protein subunits, but accurate quantification requires careful consideration of labeling efficiency and background noise.
  • - A common method for labeling involved attaching a fluorophore to cysteine residues, although its effectiveness can vary based on the protein's structure and environment.
  • - The authors developed a systematic approach to evaluate potential cysteine labeling sites on two specific membrane proteins, ultimately finding that only about 30% of the originally chosen sites were effective for their analysis despite initial hypotheses.*
