Fucosyltransferases (Fut) regulate the fucosylation process associated with tumorogenesis in different cancer types. Ascitic fluid (AF) from patients diagnosed with advanced stage of epithelial ovarian cancer (EOC) is considered as a dynamic tumor microenvironment associated with poor prognosis. Previous studies from our laboratory showed increased fucosylation in SKOV-3 and OVCAR-3, cancer-derived cell lines, when these cells were incubated with AFs derived from patients diagnosed with EOC. In the present work we studied three fucosyltransferases (Fut 2, Fut 4, and Fut 8) in SKOV-3, OVCAR-3 and CAOV-3 cell lines in combination with five different AFs from patients diagnosed with this disease, confirming that all tested AFs increased fucosylation. Then, we demonstrate that mRNAs of these three enzymes were overexpressed in the three cell lines under treatment with AFs. SKOV-3 showed the higher overexpression of Fut 2, Fut 4, and Fut 8 in comparison with the control condition. We further confirmed, in the SKOV-3 cell line, by endpoint PCR, WB, and confocal microscopy, that the three enzymes were overexpressed, being Fut 4 the most overexpressed enzyme compared to Fut 2 and Fut 8. These enzymes were concentrated in vesicular structures with a homogeneous distribution pattern throughout the cytoplasm. Moreover, we found that among the three enzymes, only Fut 4 was located inside the nuclei. The nuclear location of Fut 4 was confirmed for the three cell lines. These results allow to propose Fut 2, Fut 4, and Fut 8 as potential targets for EOC treatment or as diagnostic tools for this disease.

Cette note de recherche vise à présenter comment la science-fiction fut utilisée dans un projet de recherche pour coconstruire une vision commune de la robotique sociale favorisant la participation sociale des personnes aînées. Une recherche-action a été réalisée à l'aide de deux forums d'informateurs-clés regroupant des personnes aînées animés à partir d'extraits d'œuvres cinématographiques de science-fiction dans le but de stimuler leur réflexion. Une analyse de contenu thématique de ces forums a permis de mettre en évidence la contribution de l'usage de la science-fiction dans le cadre de cette démarche de recherche.

We present the design and synthesis of two novel isostructural metalloring organic frameworks (MROFs) {[(MeNH)(HO)NaX(μ-OH)(HO)(SIP)]·sol} (FUT-2-X, X = Eu and Sm), which are composed of the sulfonate-carboxylate ligand 5-sulfoisophthalic acid monosodium salt (NaHSIP) and an unprecedented lanthanide-sodium metalloring [XNa(HO)(SIP), X = Eu and Sm]. The two MROFs possess channel walls decorated with uncoordinated sulfonic acid groups and filled with abundant guest molecules residing within the framework, which support the proton conductivity of the materials by expanding the intermolecular hydrogen bonding network. N-glycans with complex core chitobiose modifications are observed in various free-living and parasitic nematodes but are absent in mammals. Using Caenorhabditis elegans as a model, we demonstrated that the core N-acetylglucosamine (GlcNAc) residues are modified by three fucosyltransferases (FUTs), namely FUT-1, FUT-6, and FUT-8. Interestingly, FUT-6 can only fucosylate N-glycans lacking the α1,6-mannose upper arm, indicating that a specific α-mannosidase is required to generate substrates for subsequent FUT-6 activity.
