Creating in vitro culture platforms for monkey embryos is crucial for understanding the initial 4 weeks of early primate embryogenesis. Here, we present a protocol to culture cynomolgus monkey embryos in vitro for 25 days post-fertilization and to delineate the key developmental events of gastrulation and early organogenesis. We describe steps for culturing with a 3D system, immunofluorescence analysis, single-cell RNA sequencing, and bioinformatic analysis. For complete details on the use and execution of this protocol, please refer to Gong et al. (2023)..

It has long been speculated that the mechanical properties of the human oocyte can be an indicator for oocyte viability. Recent studies have demonstrated that embryo implantation rates, following Intra-Cytoplasmic Sperm Injection (ICSI) procedures, may be increased if the shear modulus value of the oocyte Zona Pellucida (ZP) is taken into consideration during embryo transfer. The shear modulus was determined by an iterative oocyte specific finite element (FE) analysis based on the clinical ICSI data.

Cholesterol mediates the potential adverse influence of graphene quantum dots on placental lipid membrane model.

Nanomaterial-biomembrane interactions constitute a critical biological process in assessing the toxicity of such materials in theoretical studies. However, many investigations simplify these interactions by using membrane models containing only one or a few lipid types, deviating significantly from the complexity of real membrane compositions. In particular, cholesterol, a ubiquitous lipid essential for regulating membrane fluidity and closely linked to various diseases, is often overlooked.

To investigate the correlation between fetoplacental circulation and maternal left ventricular myocardial work (MW) parameters in patients with preeclampsia (PE) and the prediction of fetal hypoxia. Seventy-eight PE patients (PE group) were assigned to intrauterine-hypoxia (27) and non-intrauterine-hypoxia (51) groups, and 45 healthy pregnant women were controls. The receiver operating characteristic (ROC) curve evaluated the diagnostic efficacy of each parameter for fetal intrauterine hypoxia.

Exosomes derived from umbilical cord mesenchymal stem cells promote healing of complex perianal fistulas in rats.

National Colorectal Disease CenterNanjing Hospital of Chinese Medicine, Affiliated to Nanjing University of Chinese Medicine, Nanjing, 210022, Jiangsu, People's Republic of China.

Background: Complex perianal fistulas, challenging to treat and prone to recurrence, often require surgical intervention that may cause fecal incontinence and lower quality of life due to large surgical wounds and potential sphincter damage. Human umbilical cord-derived MSCs (hUC-MSCs) and their exosomes (hUCMSCs-Exo) may promote wound healing.

Methods: This study assessed the efficacy, mechanisms, and safety of these exosomes in treating complex perianal fistulas in SD rats.

Exploring key embryonic developmental morphokinetic parameters that affect clinical outcomes during the PGT cycle using time-lapse monitoring systems.

Research Question: Is it possible to predict blastocyst quality, embryo chromosomal ploidy, and clinical pregnancy outcome after single embryo transfer from embryo developmental morphokinetic parameters?

Design: The morphokinetic parameters of 1011 blastocysts from 227 patients undergoing preimplantation genetic testing were examined. Correlations between the morphokinetic parameters and the quality of blastocysts, chromosomal ploidy, and clinical pregnancy outcomes following the transfer of single blastocysts were retrospectively analyzed.

Results: The morphokinetic parameters of embryos in the high-quality blastocyst group were significantly shorter than those in the low-quality blastocyst group (p < 0.
