In DAF-7/TGF-beta signaling regulates development, metabolism, and behavior. In addition loss of leads to an increase of the glutamate receptor GLR-1. In mutants, GLR-1 tagged with GFP (GLR-1::GFP) accumulates in wide puncta along the ventral nerve cord of the animal. Previous automated analyses of GLR-1::GFP accumulation relied on the proprietary software, IgorPro, for measurement of GLR-1::GFP puncta size, intensity, and density. We did a side-by-side comparison of analyses by IgorPro and an open source macro written for Fiji to analyze images from animals expressing GLR-1::GFP in wild type and backgrounds. Analyses by the two programs were in strong agreement and are in accordance with previously published data on the effects of on GLR-1::GFP accumulation. Based on these data, we conclude that the Fiji platform is a robust method for analyzing the accumulation of a fluorescently-tagged neurotransmitter receptor and that the Fiji puncta plugin will be applicable for image analysis for other neural markers.

Reassessing the roles of oxidative DNA base lesion 8-oxoGua and repair enzyme OGG1 in tumorigenesis.

ROS cause multiple forms of DNA damage, and among them, 8-oxoguanine (8-oxoGua), an oxidized product of guanine, is one of the most abundant. If left unrepaired, 8-oxoGua may pair with A instead of C, leading to a mutation of G: C to T: A during DNA replication. 8-Oxoguanine DNA glycosylase 1 (OGG1) is a tailored repair enzyme that recognizes 8-oxoGua in DNA duplex and initiates the base excision repair (BER) pathway to remove the lesion and ensure the fidelity of the genome.

Background: Oocyte maturation defect (OMD) and early embryonic arrest result in female infertility. Previous studies have linked biallelic mutations in the PATL2 gene to OMD, yet the underlying mechanism remains largely unknown.

Results: This study uncovers three novel mutations (c.

This study aimed to investigate the temporal accumulation of odor fatty acids (OFAs) in the dorsal subcutaneous adipose tissue, and uncover their dynamic regulatory metabolic pathways from the transcriptomic perspective in lambs from birth to market. Thirty-two Hulun Buir lambs were selected and randomly assigned to four different sampling stages following their growth trajectories: neonatal (day 1), weaning (day 75), mid-fattening (day 150), and late-fattening (day 180) stages. Results indicated that the contents of three OFAs increased progressively as lambs matured, with the most drastic change occurred at mid-fattening vs.

Over the past few decades, microtubules have been targeted by various anticancer drugs, including paclitaxel and eribulin. Despite their promising effects, the development of drug resistance remains a challenge. We aimed to define a novel cell death mechanism that targets microtubules using eribulin and to assess its potential in overcoming eribulin resistance.
