PROteolysis TArgeting Chimeras (PROTACs) offer new opportunities in modern medicine by targeting proteins that are intractable to classic inhibitors. Heterobifunctional in nature, PROTACs are small molecules that offer a unique mechanism of protein degradation by hijacking the ubiquitin-mediated protein degradation pathway, known as the ubiquitin-proteasome system. Herein, we present an analysis on the structural characteristics of this novel chemical modality. Furthermore, we review and discuss the formulation opportunities to overcome the oral delivery challenges of PROTACs in drug discovery.

pH changes occur during bodily lesions, presenting an opportunity for leveraging pH-responsive delivery systems as signals for a targeted response. This review explores the design and application of pH-responsive delivery systems based on natural polysaccharides for the controlled release of bioactives. The article examines the development of diverse delivery carriers, including nanoparticles, nanofibers, nanogels, core-shell carriers, hydrogels, emulsions as well as liposomes and their capacity to respond to pH variations, enabling the precise and targeted delivery of bioactives within the human body.

The purpose of this review is to better characterize the contribution and properties of FDA-approved drugs that can be found unmodified in nature. Defined inclusion criteria were applied to drugs identified in previous studies and in annual FDA approval reports to compile a comprehensive list of approved drugs found in nature. Databases and scientific literature were searched to identify chemical and drug properties of these entities, including chemical classes, approval years, drug indications, and approved delivery methods.
