Reactive oxygen species (ROS) are pivotal signaling molecules that control a variety of physiological functions. As a member of the ROS family, peroxynitrite (ONOO) possesses strong oxidation and nitrification abilities. Abnormally elevated levels of ONOO can lead to cellular oxidative stress, which may cause several diseases. In this work, based on the rhodamine fluorophore, we designed and synthesized a novel small-molecule fluorescent probe () for ONOO. Upon reaction with ONOO, exhibited a significant fluorescence signal enhancement (approximately 34-fold). Moreover, showed an excellent mitochondria-targeting capability. Confocal fluorescence imaging validated its ability to detect ONOO changes in HeLa and RAW264.7 cells. Notably, we observed the ONOO generation during the ferroptosis process by taking advantage of the probe. displayed good biocompatibility, facile synthesis, and high selectivity, and may have potential applications in the study of ONOO-associated diseases in biosystems.

Compared to conventional nanocarrier-based drug delivery technology, small-molecule-assembled nanomaterials provide various advantages, including higher drug loading efficiency, lower excipient-related toxicity, and a simpler formulation process. Our research constructed a mannonse-modified small-molecule-assembled nanodrug for synergistic photodynamic/chemotherapy against A549 cancer cells. The hydrophobic hypoxic-activated agent tirapazamine (TPZ) and a hydrophilic fluorescence probe Cyanine 3 (Cy3) constitute this amphiphilic prodrug via a glutathione (GSH)-responsive linkage, which could self-assemble into stable nanoparticles (NPs) and encapsulate a newly synthesized photosensitizer (SeBDP).

Small molecules are essential for investigating the pharmacology of membrane proteins and remain the most common approach for therapeutically targeting them. However, most experimental small molecule screening methods require ligands containing radiolabels or fluorescent labels and often involve isolating proteins from their cellular environment. Additionally, most conventional screening methods are suited for identifying compounds with moderate to higher affinities ( < 1 μM) and are less effective at detecting lower affinity compounds, such as weakly binding molecular fragments.

Currently, the study of cuproptosis focuses on the Cu-induced morphology changes in mitochondria (Mito), and the observation of the effect of endoplasmic reticulum (ER)-related Cu content on cuproptosis is relatively lacking. Herein, we have developed a hydroxyflavone (HF)-based NIR excited two-photon fluorescent probe, BHCO, that exhibits specific recognition of Cu with high resolution. BHCO-Cu (Cu2BC) can lead to DLAT protein aggregation, triggering cuproptosis.
