AI Article Synopsis

  • Amyloid-β aggregation is a key feature of Alzheimer's disease, prompting research into natural compounds that can inhibit this process.
  • Recent phytochemical studies identified and purified several alkaloids and tyrosol from specific plants, finding that uncarine E and tyrosol showed significant anti-amyloidogenic effects, with inhibitions of 91% and 76% respectively at 50 µM concentrations.
  • This study represents the first known report on the anti-amyloid aggregation activity of these compounds, indicating potential for further research into their mechanisms of action against Aβ accumulation.

Article Abstract

The aggregation of amyloid-β (Aβ) remains the most acceptable pathological hallmark of Alzheimer's disease (AD). In search for natural products exhibiting anti-amyloidogenic effects, phytochemical analyses were conducted on leading to the purification of oxindole alkaloid uncarine E (isopteropodine) (), the phenylethanoid tyrosol (), the megastigmane vomifoliol (), and the previously reported alkaloids mitraphylline and isomitraphylline. This is the first report of compounds - in the title plant, and tyrosol () from the genus . Assessment of the anti-amyloidogenic potential using Thioflavin T assay showed 91% (at 50 µM) and 70% (at 25 µM) inhibitions for compound . Tyrosol () gave 76% (at 50 µM) and 63% (at 25 µM) inhibitions. These compounds may be further tested to elucidate their mechanism in the prevention of Aβ aggregation. To the best of our knowledge, this is the first report on the anti-amyloid aggregation activity of compounds and .

Nitrogen doped Carbon Quantum Dots (NCQDs) have been synthesized using most economical and easiest hydrothermal process. Here, N-phenyl orthophenylenediamine and citric acid were utilised as a source of nitrogen and carbon for the preparation of NCQDs. The synthesized NCQDs were characterized using experimental techniques like UV - Vis absorption, FT-IR, transmission electron microscopy (TEM), X-ray Diffraction (XRD), EDX, dynamic light scattering (DLS), fluorimeter and time resolved fluorescence spectroscopy.
