is a symbiont with a strong malaria transmission-blocking phenotype in . It spreads in mosquito populations through mother-to-offspring and sexual transmission. The ability of to block transmission, together with its ability to spread within populations and its avirulence to the host, makes it a very attractive candidate for developing a key strategy to stop malaria transmissions. Here, we report tissue tropism and localization patterns of , which are relevant to its transmission. We find that accumulates in tissues, linked to its sexual and vertical transmission. Its prevalence and intensity in the tissues over the mosquito life cycle suggest adaptation to maximize transmission and avirulence in . These findings provide the foundation for understanding the factors that may affect transmission efficiency. This will contribute to the development of strategies to maximize transmission to establish and sustain a high prevalence of the symbiont in mosquito populations for malaria transmission blocking.

Brain metastases (BrMets), common for advanced-stage breast cancer patients, are associated with poor median survival and accompanied by severe neurologic decline. Halting the progression of breast cancer brain metastases (BCBMs) may require modulation of the tumor microenvironment (TME), yet little is known about the impact of the primary breast TME on brain tropism, or how, once there, metastatic breast cancer cells coexist with brain-resident cells (e.g.

A Comprehensive Atlas of AAV Tropism in the Mouse.

January 2025

Department of Integrative Physiology, Baylor College of Medicine, Houston, TX 77030, USA. Electronic address:

Gene therapy with Adeno-Associated Virus (AAV) vectors requires knowledge of their tropism within the body. Here we analyze the tropism of ten naturally occurring AAV serotypes (AAV3B, AAV4, AAV5, AAV6, AAV7, AAV8, AAV9, AAVrh8, AAVrh10 and AAVrh74) following systemic delivery into male and female mice. A transgene expressing ZsGreen and Cre recombinase was used to identify transduction in a cell-dependent manner based on fluorescence.

Macrophages have emerged as promising cellular vehicles for the delivery of therapeutic agents to tumor sites. However, the cytotoxicity of therapeutic agents toward the cellular carriers and the effective release of therapeutic agents at the tumor site remain the main challenges faced by macrophage-mediated drug delivery systems. Herein, a near-infrared (NIR)-triggered release of self-accelerating cascade nanoreactor (HCFG) delivered by macrophages (HCFG@R) was developed for synergistic tumor photothermal therapy (PTT)/starvation therapy (ST)/chemodynamic therapy (CDT).

Most rare diseases (RDs) encompass a diverse group of inherited disorders that affect millions of people worldwide. A significant proportion of these diseases are driven by functional haploinsufficiency, which is caused by pathogenic genetic variants. Currently, most treatments for RDs are limited to symptom management, emphasizing the need for therapies that directly address genetic deficiencies.

Macrophage-Mediated Liquid Metal Nanoparticles for Enhanced Tumor Accumulation and Inhibition.

ACS Biomater Sci Eng

January 2025

State Key Laboratory of New Textile Materials and Advanced Processing Technologies, Wuhan Textile University, Wuhan 430200, China.

In most studies, the penetration of nanoparticles into tumors was mainly dependent on the enhanced permeability and retention (ERP) effect. However, the penetration of nanoparticles would be limited by tumor-dense structure, immune system, and other factors. To solve these problems, macrophages with active tropism to tumor tissues, loaded nanoparticles with photothermal therapy, and chemotherapy were designed.
