Preclinical xenotransplantation research using genetically engineered pigs has begun to show some promising results and could one day offer a scalable means of addressing organ shortage. While it is a fundamental tenet of ethical human subject research that participants have a right to withdraw from research once enrolled, several scholars have argued that the right to withdraw from xenotransplant research should be suspended because of the public health risks posed by xenozoonotic transmission. Here, we present a comprehensive critical evaluation of the claim that xenotransplant recipients should be required to waive their right to withdraw from lifelong biosurveillance.

Intrafemoral Injection of Human Hematopoietic Stem and Progenitor Cells into Immunocompromised Mice.

J Vis Exp

December 2023

Wellcome and Medical Research Council Cambridge Stem Cell Institute, Department of Haematology, University of Cambridge;

Hematopoietic stem cells (HSCs) are defined by their lifelong ability to produce all blood cell types. This is operationally tested by transplanting cell populations containing HSCs into syngeneic or immunocompromised mice. The size and multilineage composition of the graft is then measured over time, usually by flow cytometry.

Article Synopsis
  • The porcine cytomegalovirus (PCMV/PRV) poses a significant risk during pig-to-human transplants, as it can lead to reduced survival rates in recipients due to its ability to establish lifelong infections.
  • This study evaluated various detection methods for PCMV/PRV in donor pigs, including PCR-based techniques and immunological assays, finding that they deliver similar and reliable results.
  • Effective screening strategies combining these methods are essential to safely prevent the transmission of the virus during xenotransplant procedures.

Transplantation of porcine adrenal spheroids for the treatment of adrenal insufficiency.

Xenotransplantation

November 2023

Department of Internal Medicine III, University Hospital Carl Gustav Carus, Technical, University Dresden, Dresden, Germany.

Primary adrenal insufficiency is a life-threatening disorder, which requires lifelong hormone replacement therapy. Transplantation of xenogeneic adrenal cells is a potential alternative approach for the treatment of adrenal insufficiency. For a successful outcome of this replacement therapy, transplanted cells should provide adequate hormone secretion and respond to adrenal physiological stimuli.
