Discrimination and generalization are crucial brain-wide functions for memory and object recognition that utilize pattern separation and completion computations. Circuit mechanisms supporting these operations remain enigmatic. We show lateral entorhinal cortex glutamatergic (LEC ) and GABAergic (LEC ) projections are essential for object recognition memory. Silencing LEC during two-photon imaging increased the population of active CA3 pyramidal cells but decreased activity rates, suggesting a sparse coding function through local inhibition. Silencing LEC also decreased place cell remapping between different environments validating this circuit drives pattern separation and context discrimination. Optogenetic circuit mapping confirmed that LEC drives dominant feedforward inhibition to prevent CA3 somatic and dendritic spikes. However, conjunctively active LEC suppresses this local inhibition to disinhibit CA3 pyramidal neuron soma and selectively boost integrative output of LEC and CA3 recurrent network. LEC thus promotes pattern completion and context generalization. Indeed, without this disinhibitory input, CA3 place maps show decreased similarity between contexts. Our findings provide circuit mechanisms whereby long-range glutamatergic and GABAergic cortico-hippocampal inputs bidirectionally modulate pattern separation and completion, providing neuronal representations with a dynamic range for context discrimination and generalization.

Background: Here, we aim to develop and validate a viable prognostic nomogram model for predicting a stone-free rate of kidney stones patients based on retrospective cohort analysis.
Methods: This is a retrospective study that obtained a continuous cohort from the databases of two hospitals (General Hospital of Southern Theater Command, and Guangdong Second Provincial General Hospital), including 522 patients with kidney stones who underwent Endoscopic Combined Intrarenal Surgery (ECIRS) from January 2015 to December 2022.The characteristics of the primary cohort between the SF (stone-free) and SR (stone residue) groups were identified using single factor and multivariate logistic regression analyses.

We developed a novel next-generation sequencing-based single-nucleotide polymorphism (SNP) genotyping method for second- and third-degree kinship analysis, and designed 1144- and 2045-SNP panels using one (Set A) and two sets (Sets A and B) of primers. These SNP loci were analyzed in 120 Japanese individuals, and likelihood ratios (LRs) for kinship discrimination were calculated to evaluate the effect of number of SNP loci in simulated analysis. Likelihood evaluation was performed using DNA profiles from two individuals, namely, the unknown and one reference relative.
