Unlabelled: Treatment of [Co(CO)] with the -substituted P(C-X)P ligands (X = Br, Cl; R = Pr, Bu) bearing Y = NH and CH linkers under solvothermal conditions affords the five-coordinate Co(I) and Co(III) complexes [Co(PCP-R)(CO)] and [Co(PCP-R)X]. The later are paramagnetic exhibiting a solution magnetic moment in the range of 3.0-3.3 μ which is consistent with a d intermediate spin system corresponding to two unpaired electrons. In the case of P(C-X)P ligands (X = Br, Cl; R = Bu; Y = NH) the formation of the square planar Co(II) complex [Co(PCP-Bu)X] was favored. This complex gives rise to a magnetic moment of 1.8 μ being consistent with a d low spin system corresponding to one unpaired electron. All complexes are characterized by means of spectroscopic techniques (NMR, IR), HR-MS. Representative complexes were also characterized by X-ray crystallography.

In this study, we employed a novel fluorescent probe, RO7304924-which selectively targets cannabinoid 2 receptor (CB2R)-to assess the lateral mobility of CB2R within the plasma membrane of Chinese hamster ovary cells stably expressing a functional, untagged receptor variant. Utilizing confocal fluorescence recovery after photobleaching (FRAP), we quantified the diffusion coefficient and mobile fraction of CB2R, thereby demonstrating the efficacy of RO7304924 as an innovative tool for elucidating the dynamics of this major endocannabinoid-binding G protein-coupled receptor. Our present findings highlight the potential of combining advanced ligand-based fluorescent probes with FRAP for future investigations into the biochemical details of CB2R mobility in living cells, and its impact on receptor-dependent cellular processes.

One of the most important targets for natural killer (NK) cell-mediated therapy is the induction of natural killer group 2D ligand (NKG2D-L) expression. APTO253 is a small molecule that selectively kills acute myeloid leukemia (AML) cells, and it has been reported that APTO253 can induce Krüppel-like factor 4 (KLF4) expression and downregulate c-MYC expression. Recently, we discovered a novel role of APTO253 in modulating the NK cell response by inducing surface expression of NKG2D-Ls, especially MHC class I polypeptide-related sequence A (MICA), in AML cells.
