The methanolic extract of the leaves of Beille showed arginase inhibitory activity (40% at 100 µg/mL) and was then fractionated to obtain nine polyphenolic compounds. Their structures were elucidated on the basis of NMR spectroscopic data, and by comparison with data previously reported in the literature, as gallic acid (), 3,4-dihydroxybenzoic acid (), chlorogenic acid, (), corilagin (), cynaroside (), cosmosiin (), hyperoside () isoquercitrin ( and guajaverin (). These compounds have been evaluated as arginase inhibitors. Compounds , , and showed varying arginase inhibitory activities with IC values ranging from 102 to 302 μM. All the isolated compounds were previously identified in this species but their activities on arginase are reported here for the first time.

The aim of this study was to investigate the underlying mechanism of chrysophanol(Chr) in reducing inflammation and foam cell formation induced by oxidized low-density lipoprotein(ox-LDL) and to investigate the targets and pathways related to effects of Chr on coronary atherosclerosis, providing a theoretical basis for the development of new clinical drugs. RAW264.7 macrophages were cultured in vitro, and after determining the appropriate concentrations of Chr and ox-LDL for treating RAW264.

Myeloid-derived suppressor cells inhibit responses of T follicular helper cells during experimental Plasmodium yoelii infection.

Malaria remains a significant global public health problem. T follicular helper (Tfh) cells, a subset of CD4 T cells, have the capacity to regulate B cells, plasma cells, and antibody production, among other functions. Myeloid-derived suppressor cells (MDSCs) possess strong immunosuppressive abilities and can negatively regulate various immune responses.

Cancer immunotherapy has evolved significantly over the last decade, with therapeutics targeting the adaptive immune system showing exciting effects in clinics. Yet, the modulation of the innate immune system, particularly the tumor-associated innate immune cells which are an integral part of immune responses in cancer, remains less understood. The arginase 1 (Arg1) pathway is a pivotal metabolic pathway that tumor-associated innate immune cells exploit to create an immunosuppressive tumor microenvironment, leading to the evasion of immune surveillance.

Gastrointestinal stromal tumors regulate macrophage M2 polarization through the MIF/CXCR4 axis to immune escape.

October 2024

Department of Gastrointestinal Surgery, West China School of Medicine and West China Hospital, Sichuan University, Chengdu, Sichuan, China.

Purpose: The infiltration of immune cells and their roles of the infiltrating-immune cells in gastrointestinal stromal tumor (GIST) is still unclear. We aimed to discover the infiltration cell types and the relationship between the infiltrating-immune cells and the progression of GIST.

Experimental Design: Single-cell RNA sequencing were performed to discover types of the infiltrating-immune cells and to analyze CellChat between cells.

Article Synopsis
  • Arginase contributes to the development of an immunosuppressive environment in colorectal cancer, making the disease less responsive to immunotherapy; its activity in macrophages skews them toward a less effective M2-like polarization.
  • The study introduced a microparticle formulation (PG/ICG@MPs) that combines the natural compound piceatannol 3'-O-glucoside and indocyanine green to target tumors and modify immune responses through inhibiting arginase and enhancing nitric oxide levels.
  • Results showed that PG/ICG@MPs effectively promote the repolarization of macrophages towards the M1 phenotype and boost tumor immunity, demonstrating promising potential for combined cancer therapies that can be translated into clinical
