Nanopore direct RNA sequencing is a technology that allows sequencing for epitranscriptomic modifications with the possibility of a quantitative assessment. In the present work, pseudouridine (Ψ) was sequenced with the nanopore before and after the pH 7 bisulfite reaction that yields stable ribose adducts at C1' of Ψ. The adducted sites produced greater base call errors in the form of deletion signatures compared to Ψ. Sequencing studies on rRNA and tmRNA before and after the pH 7 bisulfite reaction demonstrated that using chemically-assisted nanopore sequencing has distinct advantages for minimization of false positives and false negatives in the data. The rRNA from has 19 known U/C sequence variations that give similar base call signatures as Ψ, and therefore, are false positives when inspecting base call data; however, these sites are refractory to reacting with bisulfite as is easily observed in nanopore data. The tmRNA has a low occupancy Ψ in a pyrimidine-rich sequence context that is called a U representing a false negative; partial occupancy by Ψ is revealed after the bisulfite reaction. In a final study, 5-methylcytidine (mC) in RNA can readily be observed after the pH 5 bisulfite reaction in which the parent C deaminates to U and the modified site does not react. This locates mC when using bisulfite-assisted nanopore direct RNA sequencing, which is otherwise challenging to observe. The advantages and challenges of the overall approach are discussed.

Plant volatile aldehydes (PVAs) such as cinnamaldehyde (Cin), citral (Cit), citronellal (Citr), and perillaldehyde (Per) have broad-spectrum antimicrobial activity and show great potential in agricultural sustainable production. However, most PVAs not only have very high volatility but also are easily degradable in environment, which seriously restricts their wide application. To address the inherent problems with PVAs, four prodrugs based on PVAs are fabricated by conjugating individually Cin, Cit, Citr, and Per to sodium bisulfite (Sod) through a simple addition reaction and subsequently self-assembled into nanoparticles (prodrug self-assemblies) in aqueous solutions.

Background: Spinal cord injury (SCI) inflicts a severe burden on patients and lacks effective treatments. Owing to the poor regenerative capabilities of endogenous oligodendrocyte precursor cells (OPCs) following SCI, there is a growing interest in alternative sources, such as human umbilical cord mesenchymal stem cells (HUCMSCs). TET3 is a key DNA demethylase that plays an important role in neural differentiation, but its role in OPC formation is not well understood.

Background: Cardiac fibrosis, characterized by excessive extracellular matrix (ECM) deposition in the myocardium, is an important target for heart disease treatments. (paternally expressed gene 3) is an imprinted gene expressed from the paternal allele, and de novo purine biosynthesis (DNPB) is a crucial pathway for nucleotide synthesis. However, the roles of PW1 and DNPB in ECM production by cardiac fibroblasts during myocardial ischemia are not yet understood.

Aiming at the problem of excessive swelling of conventional microspheres for oilfield use, a novel amphiphilic polymerizable crosslinker (AE) was synthesized by quaternary ammonium modification of an unstable crosslinker (AE) using acrylamide, 2-acrylamido-2-methylpropanesulfonic acid as the monomers, N,N'-methylene bisacrylamide as the stabilizing crosslinker, ammonium peroxysulfate and sodium bisulfite as the initiator, and water as the solvent by using a reversed microemulsion method. Double-networked nanomicrospheres were prepared. The preparation conditions of the microspheres were optimized by the surface response method, focusing on the effects of the initiator addition and reaction temperature, and total crosslinker addition on the formation of nanomicrospheres.

Introduction: Forkhead box E1 () is a transcription factor with a crucial role in thyroid morphogenesis and differentiation. Promoter hypermethylation downregulates expression in different tumor types; nevertheless, its expression and relationship with methylation status in differentiated thyroid cancer (DTC) remain unclear.
Methods: A total of 33 pairs of matched samples of PTC tumors and non-tumors were included.
