Here, we present a protocol for the simultaneous analysis of peripheral and intratumoral lymphocyte function by flow cytometry. Using tumor and blood samples from patients with colorectal cancer, we describe steps for tumor digestion, pre-labeling of the tumor-infiltrating leukocytes (TILs), and activation and labeling of the total cells (TILs and whole blood cells). For complete details on the use and execution of this protocol, please refer to Thibaudin et al..

Purpose: Intravitreal injections of anti-VEGF agents are considered as safe, with a very low rate of intraocular inflammations (IOI). Faricimab is a novel intravitreal bispecific antibody targeting both VEGF-A and angiopoietin-Tie2 independently. Despite a safe profile in randomized clinical trials, several real-life studies have reported cases of IOI.

Cluster analysis of sarcopenia in older adults: significant factors contributing to disease severity.

Federal State Autonomous Educational Institution of Higher Education "Russian National Research Medical University named after N.I. Pirogov" of the Ministry of Health of the Russian Federation, Separate structural unit "Russian Gerontology Research and Clinical Centre", 16 1st Leonova Street, Moscow, Russia, 129226.

Introduction: The European Working Group on Sarcopenia in Older People (EWGSOP2) defines sarcopenia as a muscle disease (muscle failure) rooted in adverse muscle changes that accrue across a lifetime; sarcopenia is common among adults of older age. New findings on the hormonal and metabolic characteristics of patients with sarcopenia have aided in developing more targeted therapeutic strategies. However, treating older patients with sarcopenia still poses a number of challenges.

Purpose: To evaluate image quality (IQ) of for-processing (raw) and for-presentation (clinical) radiography images, under different exposure conditions and digital image post-processing algorithms, using a phantom that enables multiple detection tasks.

Methods: A modified version of the radiography phantom described in the IAEA Human Health Series No. 39 publication was constructed, incorporating six additional Aluminum (Al) targets of thicknesses both smaller and larger than the standard one.

Single-cell sequencing in diabetic retinopathy: progress and prospects.

J Transl Med

January 2025

Department of Ophthalmology, Renmin Hospital of Wuhan University, Jiefang Road, Wuhan, Hubei, 430060, China.

Diabetic retinopathy is a major ocular complication of diabetes, characterized by progressive retinal microvascular damage and significant visual impairment in working-age adults. Traditional bulk RNA sequencing offers overall gene expression profiles but does not account for cellular heterogeneity. Single-cell RNA sequencing overcomes this limitation by providing transcriptomic data at the individual cell level and distinguishing novel cell subtypes, developmental trajectories, and intercellular communications.
