Metal-free synthesis of α-acyloxy ketones from carboxylic acids and sulfoxonium ylides.

A straightforward, catalyst- and additive-free approach has been described for synthesizing α-acyloxy ketones from β-ketosulfoxonium ylides and carboxylic acids. Moderate to high yields of α-acyloxy ketones were produced using sulfoxonium ylides and carboxylic acids adorned with various functional groups. Eventually, the applicability of this approach has been shown a large-scale reaction and transforming the synthesized α-acyloxy ketone derivatives into other valuable compounds.

Developing efficient strategies for the deoxygenative functionalization of carbonyl compounds is crucial for enhancing the effective utilization of biomass and the upgrading of chemical feedstocks. In this study, we present an elegant cathodic reduction strategy that enables a tandem alkylation/dearomatization reaction between quinoline derivatives and aryl aldehydes/ketones in a one-pot process. Our approach can be executed via two distinct paths: the aluminum (Al)-facilitated spin-center shift (SCS) path and the Al-facilitated direct deoxygenation path.

Ketone bodies generated in hepatocytes in the adult liver are used for nonhepatic tissues as an energy source. However, ketolysis is reactivated in hepatocellular carcinoma (HCC) cells with largely unelucidated mechanisms. Here, we demonstrate that 3-oxoacid CoA-transferase 1 (OXCT1), a rate-limiting enzyme in ketolysis, interacts with SUCLA2 upon IGF1 stimulation in HCC cells.

The asymmetric total synthesis of isolinearol, a -dolastane-type diterpenoid that inhibits byssal thread formation by mussels, has been achieved. In the synthesis, the key features include an intramolecular reductive nucleophilic addition using a low-valence titanium species and the direct installation of a ketone side chain. We evaluated their biological activities using the synthetic samples and found the novel inhibitory molecules with a simplified structure exhibit high inhibitory activities against byssus formation and low toxicities.
