Ovarian cancer survival depends strongly on the time of diagnosis. Detection at stage 1 must be the goal of liquid biopsies for ovarian cancer detection. We report the development and validation of graphene-based optical nanobiosensors (G-NBSs) that quantify the activities of a panel of proteases, which were selected to provide a crowd response that is specific for ovarian cancer.

Sildenafil, a phosphodiesterase-5 (PDE5) inhibitor, is widely used for erectile dysfunction and pulmonary hypertension, with its cardiovascular safety profile being well documented. However, its potential to induce conduction abnormalities remains largely unexplored, both due to a lack of clinical reports and limited mechanistic studies. While tachyarrhythmias have been frequently associated with sildenafil use, bradyarrhythmias, particularly complete heart block, are an unreported complication.

Mitochondrion apart from being the energy hub of the cell, is also the center of various signaling pathways. During intracellular infection, either bacterial or viral, several pathogen proteins, metabolites, and possibly, lipids interact with the host mitochondria. These interactions allow the pathogens, such as Mtb, to reprogram the host mitochondrial functions, screwing the host immune responses and resulting in the persistence of the bacteria.

Chronic stress is one of the potential causes of the progression of metabolic syndrome (MS). Chronic stress decreases the release of Sirtuin-6 (SIRT6), which regulates MS by controlling glucose, insulin, lipids, and hypertension. Vagus nerve stimulation (VNS) activates SIRT6 via the cholinergic anti-inflammatory pathway (CAP).

Interleukin-1 receptor-associated kinase 4 (IRAK4), a key component of the Myddosome complex, mediates signaling through toll-like and interleukin-1 receptors. KT-474, a heterobifunctional IRAK4 degrader, was evaluated in a randomized, double-blind, placebo-controlled Phase 1 trial (NCT04772885) in single (25, 75, 150, 300, 600, 1000, and 1600 mg) and multiple (25, 50, 100, and 200 mg once daily [QD] for 14 days; or 200 mg twice weekly) ascending doses in healthy subjects. The pharmacokinetics of KT-474 and its diastereomers, the pharmacodynamics of KT-474, and the effect of food on KT-474 pharmacokinetics and the pharmacokinetic-pharmacodynamic analysis are presented as additional analyses to supplement the Ackerman et al.
