Prospective isolation of defined cell types is critical for the functional study of stem cells, especially in primary human tissues. Here, we present a protocol for purifying 10 transcriptomically and functionally distinct neural stem and progenitor cell types from the developing human brain using fluorescence-activated cell sorting. We describe steps for tissue dissociation, staining, and cell sorting as well as downstream functional experiments for measuring clonogenicity, differentiation, and engraftment potential of purified populations. For complete details on the use and execution of this protocol, please refer to Liu et al. (2023)..

Early neurological deterioration in acute ischemic minor stroke patients with large vessel occlusion following intravenous thrombolysis.

World Neurosurg

December 2024

Department of Neurology, Nanjing Pukou People's Hospital, No. 166 Shanghe Street, Jiangpu Subdistrict, Pukou District, Nanjing, 210000, China. Electronic address:

Objective: Early identification of risk factors associated with early neurological deterioration (END) in patients with acute minor stroke and large vessel occlusion (LVO) receiving intravenous thrombolysis (IVT) could assist in formulating treatment decisions.

Methods: Consecutive patients with acute minor stroke and LVO were extracted from a single-center prospective database spanning January 2020 to December 2023. END was defined as an increase of ≥ 4 points in the National Institutes of Health Stroke Scale (NIHSS) score from baseline, or ≥ 2 points in any single NIHSS item, within 24 hours of the IVT.

Opportunistic screening is essential to improve the identification of individuals with osteoporosis. Our group has utilized image texture features to assess bone quality using clinical MRIs. We have previously demonstrated that greater heterogeneity of MRI texture related to history of fragility fractures, lower bone density, and worse microarchitecture.

Clinically validated classification of chronic wounds method with memristor-based cellular neural network.

Sci Rep

December 2024

Department of Electronics and Telecommunications, Politecnico di Torino, 10123, Torino, Italy.

Chronic wounds are a syndrome that affects around 4% of the world population due to several pathologies. The COV-19 pandemic has enforced the need of developing new techniques and technologies that can help clinicians to monitor the affected patients easily and reliably. In this prospective observational study a new device, the Wound Viewer, that works through a memristor-based Discrete-Time Cellular Neural Network (DT-CNN) has been developed and tested through a clinical trial of 150 patients.

Purpose: This study aimed to analyze changes in serum estradiol (E2) levels during concurrent vaginal estradiol therapy and adjuvant letrozole in postmenopausal breast cancer (BC) patients with vulvovaginal atrophy (VVA). Secondary objectives included assessing the effects of therapy on vaginal atrophy, quality of life (QoL) and menopause-related symptoms.

Methods: 20 postmenopausal patients undergoing adjuvant letrozole therapy and experiencing VVA symptoms were treated with vaginal estradiol for 12 weeks.

Pre-eclampsia (PE) is a serious condition affecting 2-8% of pregnancies worldwide, leading to high maternal and fetal morbidity and mortality. MicroRNAs (miRNAs), small non-coding RNA molecules, have emerged as potential biomarkers for various pregnancy-related pathologies, including PE. MiRNAs in plasma and serum have been extensively studied, but urinary miRNAs remain underexplored, especially during early pregnancy.
