This study addresses the propagation challenges faced by 'Shine Muscat', a newly introduced premium grapevine cultivar in South Korea, where multiple viral infections pose considerable economic loss. The primary objective was to establish a robust in vitro propagation method for producing disease-free grapes and to identify effective plant growth regulators to facilitate large-scale mass cultivation. After experimentation, 2.0 µM 6-benzyladenine (BA) exhibited superior shoot formation in the Murashige and Skoog medium compared with kinetin and thidiazuron. Conversely, α-naphthaleneacetic acid (NAA) hindered shoot growth and induced callus formation, while indole-3-butyric acid (IBA) and indole-3-acetic acid (IAA) demonstrated favorable root formation, with IBA showing better results overall. Furthermore, inter simple sequence repeat analysis confirmed the genetic stability of in vitro-cultivated seedlings using 2.0 μM BA and 1.0 μM IBA, validating the suitability of the developed propagation method for generating disease-free 'Shine Muscat' grapes. These findings offer promising prospects for commercial grape cultivation, ensuring a consistent supply of healthy grapes in the market.

Fresh grapes were stored at 4 °C and 25 °C for 28 d. The salable weight and appearance-related quality were better maintained in low-temperature storage (4 °C), and grapes stored at 25 °C presented a higher total soluble solids/titratable acidity ratio (TSS/TA), total phenols, total flavonoids, and total anthocyanins during storage compared to 4 °C.

Herein, we studied changes in the contents of phenolic and free volatile compounds in Shine Muscat grapes stored at room temperature. Berry quality was maintained up to 11 d after harvest, and the levels of 35 phenolic compounds were observed to increase during storage. This increase is attributed to the upregulation of genes, including phenylalanine ammonia-lyases, 4-coumarate-CoA ligases, and stilbene synthases, in the phenylpropanoid pathway.

High Voltage Alternating Electric Field (HVAEF), as an emerging postharvest preservation technology, is environmentally friendly and has garnered significant attention from scholars. This study investigated the effects of HVAEF on the postharvest quality and metabolite changes in 'Shine Muscat' grapes stored at near-freezing temperatures. The results demonstrated that HVAEF halved the weight loss and decay rate, inhibited browning by preserving polyphenol content, and reduced polyphenol oxidase and peroxidase activities, thereby extending the storage time by more than 20 days.
