PRMT5 contributes to secondary metabolite biosynthesis in . However, the mechanism through which PRMT5 regulates the biosynthesis of secondary metabolites remains unclear. In the current study, silencing led to a significant decrease in the biosynthesis of polysaccharides from through the action of the alternative splicing of . A shorter isoform can directly bind to PGI and regulated polysaccharide biosynthesis. These results suggest that PRMT5 enhances PGI activity by regulating TLP binding to PGI. The results of the current study reveal a novel target gene for PRMT5-mediated alternative splicing and provide a reference for the identification of PRMT5 regulatory target genes.

Protein arginine methyltransferase 5 (PRMT5) is an enzyme that produces monomethyl arginine (MMA) and symmetric dimethyl arginine (sDMA), post-translational modifications that regulate several cellular processes, including stage conversion in parasitic protozoans. , the etiologic agent of human amebiasis, has two stages in its life cycle, the trophozoite, which is the replicative form, and the cyst, corresponding to the infective phase. The study of the molecular mechanisms that regulate differentiation in this parasite has been overdue because of a lack of efficient protocols for in vitro encystment.

Protein arginine methyltransferase 5 (PRMT5), which catalyzes the symmetric dimethylation of arginine residues on target proteins, plays a critical role in gene expression regulation, RNA processing, and signal transduction. Aberrant PRMT5 activity has been implicated in cancers and other diseases, making it a potential therapeutic target. Here, we report the discovery of a methylthioadenosine (MTA) cooperative PRMT5 inhibitor.

Numerous diseases have been connected to protein arginine methylations mediated by protein arginine methyltransferase 5 (PRMT5). Clinical investigations of the PRMT5-specific inhibitor GSK3326595 are currently being conducted, and the results are promising for preventing cancers. However, the detailed mechanism of PRMT5 promoting colorectal cancer (CRC) malignant progression remains unclear.

Protein arginine methyltransferase 5 (PRMT5) is a promising cancer target, yet it's unclear which PRMT5 roles underlie this vulnerability. Here, we establish that PRMT5 inhibition induces a special class of unspliced introns, called detained introns (DIs). To interrogate the impact of DIs, we depleted CLNS1A, a PRMT5 cofactor that specifically enables Sm protein methylation.

Article Synopsis
  • TNG908 is a clinical-stage inhibitor targeting PRMT5, utilizing a unique binding mechanism that exploits the loss of the MTAP gene commonly found in various cancers.
  • It specifically inhibits PRMT5 in cancer cells lacking MTAP, which occurs in 10-15% of human cancers and could lead to more effective treatments compared to earlier drugs.
  • Ongoing Phase I/II trials are investigating the effectiveness of TNG908 in patients with MTAP-null tumors, including glioblastoma, suggesting a promising future for this therapy in multiple cancer types, particularly those affecting the brain.
